Design of a multi-epitope Zika virus vaccine candidate - an in-silico study

Summary: In this study, we designed a multi-epitope-based subunit vaccine against Zika virus using an in silico approach, and assessed its immunogenicity and stability through immunoinformatics, molecular docking, and dynamic simulation. The vaccine showed strong immune responses and stable molecular interactions with immune receptors. In addition, the vaccine was successfully cloned in E.coli using in silico methods. However, experimental validation is still needed.

Identification of B-cell epitopes on structural proteins VP1 and VP2 of Senecavirus A and development of a multi-epitope recombinant protein vaccine

Summary: B-cell epitopes of Senecavirus A (SVA) were identified using a bioinformatics approach combined with an overlapping synthetic polypeptide method. Four dominant B-cell epitopes from the VP1 protein and five dominant B-cell epitopes from the VP2 protein were identified. The multi-epitope recombinant protein rP2 induced higher neutralizing antibodies and provided 80% protection against homologous SVA challenge, showing its potential as a candidate for SVA vaccine development.

Novel Epitopes Mapping of African Swine Fever Virus CP312R Protein Using Monoclonal Antibodies

Summary: African Swine Fever (ASF), a highly contagious and lethal pig disease, poses a significant threat to the pig industry worldwide. In this study, the immunogenicity of ASFV CP312R protein was verified in pigs. Seven mouse monoclonal antibodies (mAbs) were generated, and two antigenic determinant regions of CP312R protein were identified. These findings enhance our understanding of CP312R protein and contribute to the development of serological diagnosis and vaccines for ASF.

Summary: The nonspecific enrichment of target-unrelated peptides during biopanning is a major issue in phage display technology. A fast-propagating white clone was isolated in this study through screening against a recombinant protein. It was found that this white clone was not contamination but derived from the library itself. Whole genome sequencing revealed a large genomic deletion as the cause of the white color of the plaques. Phenotypic characterization demonstrated a higher propagation rate of the white clone compared to the library. The data suggests that white plaques may arise endogenously in Ph.D. libraries due to mutations and should not always be considered as exogenous contamination.
