Scaffold geometry modulation of mechanotransduction and its influence on epigenetics

Pingping Han et al.

Summary: The dynamics of cell mechanics and epigenetic signatures play a crucial role in cell behavior and fate, affecting the outcomes of tissue regeneration. While 2D geometric substrates have been widely used to study cell mechanics in response to the extracellular microenvironment, there is limited knowledge about cell mechanobiology and epigenetics in 3D biomaterial matrices. This review explores the potential of additive manufacturing for incorporating multi length-scale geometry features on scaffolds, and discusses how scaffold geometry influences cell and nuclear mechanosensing and epigenetic modifications, ultimately impacting tissue regeneration outcomes.

Tubular Bioartificial Organs: From Physiological Requirements to Fabrication Processes and Resulting Properties. A Critical Review

Nele Pien et al.

Summary: This featured review manuscript provides a critical survey on the fabrication processes of bioartificial organs (BAOs), specifically focusing on hollow tubular organs. The manuscript presents an overview of the anatomical structures and physiological functions of tubular organs, which are essential for designing and developing physiologically relevant BAOs. Various techniques such as 3D bioprinting, solution electrospinning, and melt electrowriting are introduced, highlighting their compatibility with the anatomical and physiological environment. The manuscript also discusses the intrinsic correlation between processes, materials, and cells, providing guidelines for bioengineers in selecting the appropriate process for target BAOs and their specific functions.

Biodegradable polyurethane scaffolds in regenerative medicine: Clinical translation review

Drake D. Pedersen et al.

Summary: Early explorations of tissue engineering and regenerative medicine commonly used simple polyesters as scaffolds. However, with the advancement of technology, a greater variety of synthetic biomaterials, such as biodegradable polyurethanes, have been employed. These polyurethanes can be customized based on their composition and processability, and can incorporate bioactive agents to achieve desired biological responses. While there is much enthusiasm and growth in the literature on biodegradable PU scaffolds, their regulatory approval and routine clinical use are still limited. Nonetheless, there is a growing body of research on the development and pre-clinical applications of these scaffolds.

Effects of the 3DP process parameters on mechanical properties of polylactic acid part used for medical purposes

Sunthorn Chaitat et al.

Summary: This study investigated the effects of infill density, built orientation, and gamma radiation dose on the mechanical properties of PLA parts fabricated using FDM for medical applications. The results showed that compressive mechanical properties are generally greater than tensile mechanical properties. Increasing the number of layers parallel to the loading direction and infill density can enhance material properties. Upright orientation should be avoided for parts subjected to tensile loads, while FDM is more suitable for parts subjected to compressive loads. The findings are valuable for selecting FDM parameters in medical device manufacturing.

Current status of xenotransplantation research and the strategies for preventing xenograft rejection

Qiao Zhou et al.

Summary: Transplantation is a last resort for end-stage organ failures, but the shortage of donor organs limits its success. Xenotransplantation of pig organs is an alternative, but immune rejection is a major challenge. This review comprehensively discusses the mechanisms of immune rejection and strategies to prevent it, such as gene knockout donors and immunosuppressive regimens.
