NCAPD2 promotes breast cancer progression through E2F1 transcriptional regulation of CDK1

Jinsong He et al.

Summary: This study investigated the clinical significance, biological behavior, and potential molecular mechanism of Non-SMC condensin I complex subunit D2 (NCAPD2) in breast cancer (BC). The findings showed that NCAPD2 was frequently overexpressed in BC and had clinical significance in predicting patient prognosis. Knocking down NCAPD2 inhibited BC progression through inhibiting proliferation and migration and enhancing apoptosis in vitro. Moreover, NCAPD2 downregulation inhibited tumor growth in vivo and promoted BC progression through the ERK5 signaling pathway.

Topoisomerase IIA in adult NSCs regulates SVZ neurogenesis by transcriptional activation of Usp37

Shangyao Qin et al.

Summary: This study reveals the preferential expression of TOP2a in neurogenic niches and its crucial role in the self-renewal and proliferation of adult neural stem cells. The researchers identified Usp37 as a direct target gene of TOP2a through RNA sequencing and chromatin immunoprecipitation sequencing, and found that overexpression of Usp37 can rescue the impaired self-renewal ability of adult neural stem cells caused by Top2a knockout.

ROS Dynamics Delineate Functional States of Hippocampal Neural Stem Cells and Link to Their Activity-Dependent Exit from Quiescence

Vijay S. Adusumilli et al.

Summary: The study discovered that quiescent neural precursor cells (NPCs) in the hippocampus of adult mice maintain the highest levels of intracellular reactive oxygen species (ROS). Different functional states of NPCs were identified based on their ROS content, and physical activity was found to recruit NPCs with high ROS levels into proliferation. This mechanism provides a link between the modulation of cellular ROS by behavioral cues and the activation of adult NPCs.

TC10, a Rho family GTPase, is required for efficient axon regeneration in a neuron-autonomous manner

Shingo Koinuma et al.

Summary: Results suggest that TC10 plays a crucial role in axon regeneration within both peripheral and central nervous systems. Lack of TC10 significantly suppresses axon regeneration post-injury, while not affecting brain size and structure during development. TC10 exerts its effects on axon regeneration by stabilizing microtubules and acting as a neuron-intrinsic factor.

Shalini Dogra et al.

Summary: Activation of mGlu3 receptors in hippocampal pyramidal cells enhances cognition in both control and impaired mice by inducing a novel form of metaplasticity to regulate circuit function, and reveals a mechanism by which mGlu3 and mGlu5 interact to enhance cognitive function.

Coordinated changes in cellular behavior ensure the lifelong maintenance of the hippocampal stem cell population

Lachlan Harris et al.

Summary: The stabilization of neural stem cell numbers in young adults is a result of coordinated changes in stem cell behavior, with resting cells having a higher activation rate and greater contribution to neurogenesis. This change is associated with a progressive reduction in expression of the pro-activation protein ASCL1 due to increased post-translational degradation, providing insights into hippocampal NSC dynamics and neurogenesis rates in different mammalian species, including humans.

Neurexins regulate presynaptic GABAB-receptors at central synapses

Fujun Luo et al.

Summary: This study shows that neurexins are essential for the localization and function of presynaptic GABA(B)-receptor signaling complexes. Deletion of neurexins results in decreased sensitivity of neurotransmitter release to GABA(B)-receptor activation and loss of GABA(B)-receptors from the presynaptic active zone. Neurexins play a crucial role in regulating presynaptic GABA(B) receptors at multiple central nervous system synapses.
