Bidirectional Mendelian Randomisation Analysis Provides Evidence for the Causal Involvement of Dysregulation of CXCL9, CCL11 and CASP8 in the Pathogenesis of Ulcerative Colitis

Summary: This study identified 14 proteins associated with the risk of ulcerative colitis (UC) through the analysis of serum proteomic profiling data. Further analysis using Mendelian randomisation confirmed the causal relationship between two chemokines and the increased risk of UC. Additionally, a new protein associated with UC risk was identified. Reverse Mendelian randomisation analysis did not find any influence of genetic predisposition to UC on these three inflammation proteins.

Pyroptosis-Related Signature Predicts the Progression of Ulcerative Colitis and Colitis-Associated Colorectal Cancer as well as the Anti-TNF Therapeutic Response

Summary: Ulcerative colitis (UC) is a complex intestinal inflammation with an increasing risk of colitis-associated colorectal cancer (CAC). Pyroptosis-related genes (PRGs) have been found to be closely associated with inflammatory disease activity in UC. In this study, researchers identified differentially expressed PRGs and established a pyroptosis-related score (PR-Score) that can distinguish active UC from inactive UC and predict treatment response. The PR-Score is also correlated with immune cell infiltration, inflammatory microenvironment, and survival outcomes in colorectal cancer. These findings contribute to our understanding of UC and provide new markers for diagnosis and treatment.

Key genes such as CXCL9, C3AR1, IL1B, and TLR4 were identified as potential predictors of CD activation.

Serum biomarkers confirming stable remission in inflammatory bowel disease

Summary: Crohn's disease (CD) and ulcerative colitis (UC) have a chronic-remittent course. Optimal management of inflammatory bowel diseases (IBD) relies on early intervention, treat-to-target strategies and a tight disease control. However, it is challenging to assess the risk of relapses in individual patients. We investigated blood-based biomarkers for the confirmation of disease remission in patients with IBD. Our study identified serum biomarkers with the potential to indicate unstable disease control in IBD, which may help to steer individual therapies to maintain remission.
