Temporal and thermal profiling of the Toxoplasma proteome implicates parasite Protein Phosphatase 1 in the regulation of Ca2+-responsive pathways

Alice L. Herneisen et al.

Summary: Apicomplexan parasites utilize Ca2+ signaling pathways to regulate specific cellular processes and contribute to the pathogenesis. Mapping the Ca2+-responsive proteome of Taxoplasma gondii revealed novel proteins involved in the signaling network and identified the Ca2+-responsive enzyme PP1 as a regulator of parasite motility.

A positive feedback loop mediates crosstalk between calcium, cyclic nucleotide and lipid signalling in calcium-induced Toxoplasma gondii egress

Stephanie D. Nofal et al.

Summary: In the lytic cycle of Toxoplasma gondii, calcium and cyclic nucleotide signaling pathways are connected by a positive feedback loop, which is regulated by a plant-like calcium dependent kinase (CDPK3) and a cAMP-specific phosphodiesterase (PDE2). This feedback loop allows the parasite to respond faster to egress inducers.

PI(4,5)P2 Clustering and Its Impact on Biological Functions

Yi Wen et al.

Summary: Phosphatidylinositol 4,5-bisphosphate (PI(4,5)P-2) is a crucial component located at the inner leaflet of the cell membrane, constituting only 1-2% of total membrane lipids. Its synthesis and turnover are spatially and temporally regulated. Various factors, such as multivalent cations and cellular proteins interacting with PI(4,5)P-2, contribute to its versatile and dynamic distribution within membranes.

Protein kinase TgCDPK7 regulates vesicular trafficking and phospholipid synthesis in Toxoplasma gondii

Priyanka Bansal et al.

Summary: In this study, we demonstrate that the protein kinase TgCDPK7 regulates cellular processes such as vesicular trafficking and phospholipid synthesis, critical for the development of the parasite Toxoplasma gondii. It phosphorylates the small GTPase TgRab11a to regulate the trafficking of important parasite proteins, crucial for parasite division. TgCDPK7 may also regulate key enzymes involved in the biogenesis of phosphatidylethanolamine, contributing to the synthesis of this important phospholipid. These findings shed light on a novel signaling pathway in apicomplexan parasite Toxoplasma gondii.

Rab5b-Associated Arf1 GTPase Regulates Export of N-Myristoylated Adenylate Kinase 2 From the Endoplasmic Reticulum in Plasmodium falciparum

Izumi Taku et al.

Summary: Plasmodium falciparum extensively remodels human erythrocytes by exporting hundreds of parasite proteins, with PfRab5b regulating the transport pathways of PfAK2 in conjunction with PfArf1 and PfRab1b at different ER subdomains. This study demonstrates that Rifin and PfAK2 export are recognized at different ER subdomains by PfArf1 and PfRab1b, respectively.

Plasmodium falciparum Atg18 localizes to the food vacuole via interaction with the multi-drug resistance protein 1 and phosphatidylinositol 3-phosphate

Renu Sudhakar et al.

Summary: Autophagy, a degradative process dependent on lysosomes, does not play a major role in malaria parasites. Plasmodium falciparum Atg18 (PfAtg18) binds PI3P for localization to the food vacuole, with the multi-drug resistance protein 1 (MDR1) also interacting with PfAtg18 for localization. This indicates that PI3P and MDR1 are critical for PfAtg18 localization in the parasite.

Carolyn M. Highland et al.

Summary: Proper function of the Golgi complex depends on the activity of Arf1, which controls PI4P production to initiate the final stages of Golgi maturation, facilitating AP-1 and secretory vesicle formation. This study reveals the crucial roles of Arf1 and PI4P in Golgi organelle function and maturation process.

Emerging Prospects for Combating Fungal Infections by Targeting Phosphatidylinositol Transfer Proteins

Danish Khan et al.

Summary: The emergence of fungal superbugs resistant to current anti-fungal agents is diminishing our ability to treat infections effectively, highlighting the urgent need for new classes of anti-fungal drugs and targets. Recent evidence suggests that Sec14-like phosphatidylinositol transfer proteins (PITPs) could be unexplored portals for selectively disrupting phosphoinositide signaling in virulent fungi, offering exciting potential for next generation Sec14 inhibitors. Development of refined Sec14-directed inhibitors could enhance the chemical weaponry available for combating fungal pathogens and their interactions with human hosts.
