Regulation of insulin secretion by the post-translational modifications

Palmitoylation couples insulin hypersecretion with (3 cell failure in diabetes

Summary: Hyperinsulinemia often occurs before type 2 diabetes. The protein modification process called palmitoylation, which is involved in insulin secretion, can be reversed by APT1. Changes in APT1 function were observed in pancreatic islets of type 2 diabetes patients, and reducing APT1 levels in non-diabetic islets resulted in excessive insulin secretion. Knockout mice lacking APT1 showed increased glucose-stimulated insulin secretion in islets, along with prolonged insulin granule fusion. APT1 was found to regulate the palmitoylation of Scamp1, a protein localized in insulin secretory granules, and knocking down Scamp1 also caused insulin hypersecretion. These findings suggest that APT1 plays a role in the regulation of insulin secretion and its deficiency can lead to beta cell dysfunction, resembling the development of certain forms of human type 2 diabetes.

Understanding the mechanisms of PTMs in metabolic diseases will provide new insights for therapeutic intervention and improvement of therapies.

The dynamic clustering of insulin receptor underlies its signaling and is disrupted in insulin resistance

Insulin stimulation promotes further incorporation of IR into these clusters, while in insulin-resistant cells, IR accumulation and dynamic behavior are reduced. Treatment with the commonly used drug metformin, which reduces reactive oxygen species, can rescue IR accumulation and dynamic behavior in insulin-resistant cells. These findings have implications for improved therapeutic approaches.
