Summary: In this study, a single-cell strategy was developed to define the HIV-infected memory CD4(+) T cell reservoir in ART-treated people living with HIV (ART-PLWH) using integrated accessible proviral DNA, epigenetic and cell surface protein profiling. The study identified a profound reservoir heterogeneity within and between ART-PLWH, characterized by new and known surface markers within memory CD4(+) T cell subsets. Furthermore, new epigenetic profiles and transcription factor motifs enriched in HIV-infected cells were uncovered, suggesting a potential for reactivation during ART treatment. These findings establish an initial multiomic atlas for targeted reservoir elimination strategies.

Summary: HIV-1 infection leads to the formation of a long-lasting reservoir of latently infected CD4(+) T cells, consisting of intact and defective proviruses. This study used Q4PCR to quantify the reservoir content and found that the defective reservoir remained stable while the intact proviral reservoir decayed over time. Both the intact and defective proviral reservoirs showed dynamic changes, with an enrichment of intact proviruses in expanded CD4(+) T cell clones and a decrease in overall reservoir complexity.

Gag p24 Is a Marker of Human Immunodeficiency Virus Expression in Tissues and Correlates With Immune Response

Summary: The study demonstrates that the detection of the HIV gag p24 protein is more prominent in gut and lymph node tissues compared to blood CD4(+) T cells, and it correlates better with CD4 count. Levels of gut p24 also decrease measurably during antiretroviral therapy and are strongly associated with HIV-specific CD8(+) T-cell frequency and plasma soluble CD14 levels. Using gag p24 as a marker for HIV expression in HIV+ tissues is supported as a way to study viral persistence effects and monitor treatment efficacy in HIV-based clearance studies.
