Lactic acid promotes PD-1 expression in regulatory T cells in highly glycolytic tumor microenvironments

Summary: In highly glycolytic tumors, regulatory T (Treg) cells have higher PD-1 expression compared to effector T cells. Under low-glucose conditions, Treg cells actively absorb lactic acid through monocarboxylate transporter 1 (MCT1), promoting the expression of PD-1, while PD-1 expression by effector T cells is dampened. PD-1 blockade invigorates PD-1-expressing Treg cells, resulting in treatment failure.

LDH may play a key role in the clinical management of these diseases, and strategies targeting LDH show promising results as a treatment strategy in combination with current/novel chemotherapeutics.

Summary: In the tumor microenvironment, senescent cancer-associated fibroblasts (CAFs) play a role in promoting gastric cancer (GC) peritoneal tumor formation through maintenance of the senescence-associated secretory phenotype (SASP) and JAK/STAT3 signaling. Single-cell mass cytometry analysis revealed a high expression level of senescent fibroblasts and SASP factors in the ascites of GC patients. These findings provide insights into the molecular mechanisms of inflammation-related SASP maintenance and the involvement of senescent CAFs in GC peritoneal dissemination.

Summary: The CD155/TIGIT axis can be utilized for immune evasion in chronic viral infections and cancers like pancreatic adenocarcinoma. Studies have shown that a significant portion of PDAC patients have predicted high-affinity MHC class I-restricted neoepitopes, and modulation of the CD155/TIGIT axis can promote immune evasion in PDAC. In preclinical models, a combination immunotherapy targeting TIGIT/PD-1 plus CD40 agonism has shown promising anti-tumor responses.
