Extracellular Vesicles and Their Zeta Potential as Future Markers Associated with Nutrition and Molecular Biomarkers in Breast Cancer

Summary: A nutritional intervention was found to promote body and visceral fat loss while maintaining muscle mass in breast cancer patients. The changes in EVs' Zeta potential, miRNA-30, miRNA-145, and miRNA-155 content, and their association with body composition and metabolic risk biomarkers were explored in breast cancer patients before and after a nutritional intervention. The expression of miRNA-30 and miRNA-155 and the Zeta potential of EVs were significantly associated with biomarkers of metabolic risk and disease prognosis.

Highlights from the past decade and future perspectives for this research field are presented, outlining challenges to be addressed in the coming years.

In this study, a modified isolation method was developed to improve EV isolation and enrich prostate cancer-associated EV-resident proteins.

High-fat diet-induced upregulation of exosomal phosphatidylcholine contributes to insulin resistance

Summary: This study reveals that lean mice become insulin resistant after being administered exosomes isolated from the feces of obese mice fed a high-fat diet or from patients with type II diabetes. The altered lipid composition of exosomes from high-fat diet-fed mice affects the uptake by macrophages and hepatocytes, leading to inhibition of the insulin signaling pathway. Exosome-derived phosphatidylcholine activates AhR, inhibiting the expression of genes essential for activation of the insulin signaling pathway.

Extracellular vesicles (EVs), especially SASP-related EV signaling, play a critical role in aging transmission, facilitating intercellular and interorgan communication in the aging microenvironment while also potentially causing detrimental effects on downstream targets. EVs from different donors are promising materials for antiaging treatments, used for regeneration and rejuvenation in cell-free systems.

Reversing the surface charge of MSC-derived small extracellular vesicles by εPL-PEG-DSPE for enhanced osteoarthritis treatment

Summary: This study developed a strategy to reverse the surface charge of MSC-sEVs by modifying them with a novel cationic amphiphilic macromolecule, resulting in improved distribution and bioavailability in cartilage. The modified PPD-sEVs demonstrated enhanced cellular uptake and homeostasis modulation ability to chondrocytes, as well as significantly improved cartilage uptake, penetration, and joint retention capacity in a mouse OA model. Intra-articular injection of PPD-sEVs showed enhanced bioavailability and therapeutic efficacy compared to unmodified MSC-sEVs.

Summary: This study focused on extracellular vesicles as an important source of tumor-derived DNA in liquid biopsies, and conducted experiments by collecting blood samples from 17 pancreatic cancer patients. The results showed that tumor-derived DNA was mainly present in LEVs and SEVs in the early stage of disease progression, and in SEVs and SP in the late stage of disease progression.

Summary: The study found that during chronic methamphetamine (MA) exposure, brain-derived EV (BDE) miRNA miR-29a-3p levels significantly increased and were associated with drug-seeking and reinstatement in MA. EV-associated miR-29a was enriched in the EV pool and played a critical role in MA-induced inflammation and synaptodendritic damage. EV-miR29a could potentially serve as a biomarker to detect neuronal damage in MUD individuals.
