S-Nitrosylation of p62 Inhibits Autophagic Flux to Promote α-Synuclein Secretion and Spread in Parkinson's Disease and Lewy Body Dementia

Chang-Ki Oh et al.

Summary: This article investigates the mechanism by which dysregulation of autophagic pathways leads to the accumulation of abnormal proteins and damaged organdies in neurodegenerative disorders. The authors found that pathologic protein S-nitrosylation of p62 is a critical factor for autophagic inhibition and cell-to-cell spread.

Stimuli-Mediated Specific Isolation of Exosomes from Blood Plasma for High-Throughput Profiling of Cancer Biomarkers

Xuehui Liu et al.

Summary: This study presents the first stimuli-mediated enrichment and purification system for selectively and efficiently extracting exosomes from clinical plasma, for high-throughput profiling of exosomal mRNAs as cancer biomarkers. The system outperforms conventional methods in terms of isolation yield, purity, and retained bioactivity, demonstrating high clinical potential in cancer diagnosis and discrimination. This novel isolation system has the potential to advance extracellular vesicle research in both basic and clinical settings.

Lipid-based strategies used to identify extracellular vesicles in flow cytometry can be confounded by lipoproteins: Evaluations of annexin V, lactadherin, and detergent lysis

Jaco Botha et al.

Summary: Flow cytometry is commonly used to characterize extracellular vesicles (EVs). However, this study found that PS labeling and detergent lysis methods may be confounded by lipoproteins. It suggests using transmembrane proteins to differentiate EVs from lipoproteins.

Serpin-loaded extracellular vesicles promote tissue repair in a mouse model of impaired wound healing

Dong Jun Park et al.

Summary: Chronic metabolic diseases such as diabetes is associated with delayed wound healing and dysregulated inflammatory phase of wound repair. This study focuses on the changes in extracellular vesicles (EVs) payload that mediates communication between immune cells and stromal cells in the wound bed, which regulate the rate of wound closure. By transferring EVs from genetically defined mouse models, the functional and molecular basis for differences in the pro-reparative biological activity of diabetic (db/db) vs. wildtype EVs in wound healing is demonstrated. This study establishes a novel therapeutic approach by engineering the payload of EVs, aiming to rescue the Serpin deficiency identified by proteomics and promote wound healing in db/db mice.

Refractive Index Determination of Individual Viruses and Small Extracellular Vesicles in Aqueous Media Using Nano-Flow Cytometry

Ye Tian et al.

Summary: In this study, a label-free technique was developed to determine the refractive index (RI) of individual viruses and small extracellular vesicles (sEVs) using high-sensitivity light-scattering detection and Mie theory calculation. The RI measurements allowed for the estimation of nucleic acid concentrations in viral particles and protein concentrations in the lumen of sEVs, as well as the size distribution of sEVs.

Sizing Extracellular Vesicles Using Membrane Dyes and a Single Molecule-Sensitive Flow Analyzer

Luca A. Andronico et al.

Summary: The study demonstrates a method for sizing EVs using a fluorescence-based flow analyzer with single-molecule sensitivity. After optimizing staining conditions, accurate measurements of vesicle sizes were obtained, covering the spectrum of EV sizes. A method for correcting signal intensity based on vesicle speed inside a microfluidic channel was developed, resulting in a threefold greater accuracy in EV sizing.

Characterization of Extracellular Vesicles Labelled with a Lipophilic Dye Using Fluorescence Nanoparticle Tracking Analysis

Getnet Midekessa et al.

Summary: Research has shown that the concentration of CMG fluorescent dye affects the size of fluorescent nanoparticles (fl-NPs), with an increase in dye concentration resulting in a significant decrease in size. Zeta potential (ZP) values of fl-NPs from JAr cells also shift significantly towards more negative or less negative values with varying dye concentrations. Furthermore, the proportion of CMG labeling of NPs varies across EVs purified using different methods and from different biological sources.

Generation and Application of a Reporter Cell Line for the Quantitative Screen of Extracellular Vesicle Release

Jonathan Shpigelman et al.

Summary: Extracellular vesicles (EVs) play a key role in intercellular communication and cellular regulation, particularly in the immune system. A new reporter cell line was developed to quantitate EVs released by immune cells, enabling high-throughput screening for drug discovery and compound characterization. This study demonstrates the reliability, robustness, and feasibility of using CD63Tluc-CD9EmGFP reporter cells for HTS of compounds regulating EV release by immune cells.
