Emerging therapeutics agents and recent advances in drug repurposing for Alzheimer?s disease

Suhel Parvez Neha

Summary: Alzheimer's disease is a complex neurodegenerative disease that affects the cerebral cortex and hippocampus, leading to memory and cognitive deficits. The accumulation of amyloid beta peptides and dysfunctioning of tau proteins are the key pathological features. This review highlights the simplicity of finding novel treatments for AD, focusing on repurposed medications that can speed up drug development.

Exploiting machine learning models to identify novel Alzheimer's disease biomarkers and potential targets

Hind Alamro et al.

Summary: Despite being the most common cause of dementia and impaired cognitive function, an effective treatment for Alzheimer's disease (AD) remains elusive. This study developed a computational method that combines multiple hub gene ranking methods, feature selection methods, and machine learning to identify biomarkers and targets for AD. The results showed that feature selection methods outperformed hub gene sets in prediction performance, and a small number of genes were able to accurately distinguish AD samples from healthy controls.

Transcriptomic analysis of glutamate-induced HT22 neurotoxicity as a model for screening anti-Alzheimer's drugs

Anchalee Prasansuklab et al.

Summary: This study provides the first transcriptomic and network analyses of HT22 cells following glutamate exposure, identifying differentially expressed genes (DEGs) and their relationships specific to Alzheimer's disease (AD). The usefulness of this cell model as a drug screening system was assessed by determining the expression of AD-associated DEGs in response to two medicinal plant extracts, Acanthus ebracteatus and Streblus asper.

2023 Alzheimer's disease facts and figures

[Anonymous]

Summary: This article discusses the impact of Alzheimer's disease on public health, including its prevalence and incidence, mortality and morbidity, care usage and costs, and the effects on family caregivers, the dementia workforce, and society. It highlights the patient journey and potential treatment options for Alzheimer's disease. Currently, there are 6.7 million older Americans living with Alzheimer's dementia, a number that could increase to 13.8 million by 2060 without medical breakthroughs. Alzheimer's is the sixth-leading cause of death in the US, with deaths increasing by over 145% between 2000 and 2019. There is a decline in the number of caregivers and an increase in care provided by each remaining caregiver. The costs of unpaid caregiving in 2022 reached $339.5 billion, impacting caregivers' mental and physical health. Additionally, there is a shortage of healthcare workers in dementia care, and Medicare and Medicaid payments are significantly higher for individuals with dementia. The article also explores the need for sufficient physician specialists in Alzheimer's care, considering the availability of drugs that change the underlying biology of the disease.

I2 imidazoline receptor modulation protects aged SAMP8 mice against cognitive decline by suppressing the calcineurin pathway

Foteini Vasilopoulou et al.

Summary: By orally administering the specific and selective I-2-IR ligand B06 to SAMP8 mice, behavioral and cognitive improvements were observed, along with reduction in AD hallmarks, oxidative stress, and apoptotic and neuroinflammatory markers. B06 also regulated protein levels of glial excitatory amino acid transporter 2 and N-methyl-D-aspartate 2A and 2B receptor subunits, modulating the CaN pathway and affecting various molecular pathways to play a neuroprotective role in aging and AD.
