Optimization and in vivo evaluation of triamcinolone acetonide loaded in situ gel prepared using reacted tamarind seed xyloglucan and kappa-carrageenan for ocular delivery

Summary: In this study, a dual responsive in situ gelling system loaded with triamcinolone acetonide (TAA) was designed and optimized using reacted tamarind seed xyloglucan (RXG) and kappa-Carrageenan (kappa-CRG) polymers. The optimized formulation exhibited good flow properties at 25 degrees C but transformed into a stronger gel at 35 degrees C in the presence of STF. The ex vivo ocular toxicity studies indicated that the optimized formulation was well tolerated.

Summary: The study fabricated chitosan/hyaluronan nanogels to co-load methotrexate (MTX) and 5-aminoleavulinic acid (ALA) for a combined chemo-photodynamic therapy for psoriasis. The nanogels enhanced the penetration and retention of MTX and ALA in the skin, leading to synergistic anti-proliferative and apoptotic effects on psoriatic cells. Importantly, the nanogels reduced the toxicities of oral MTX to the liver and kidney, making it a convenient, effective, and safe treatment strategy for psoriasis.

iota-Carrageenan hydrogels for methotrexate delivery

It was found that complexation with β-cyclodextrin affects the release rate and mechanism of methotrexate, as well as the permeability of the drug through the skin. The prepared hydrogel can be proposed as a topical drug delivery system for the treatment of skin cancer and autoimmune diseases.
