Investigation of the adsorption and release kinetics of the anticancer drug, methotrexate, from chitosan nanocapsules modified by caffeic acid and oleic acid

Summary: This study compared the effects of different fatty acid-modified chitosan surfaces as nanocarriers for targeted delivery of the anticancer drug methotrexate (MTX) in vitro. The surface modification of chitosan with oleic acid (OA) and caffeic acid (CA) was investigated. The results showed that CS-CA had a higher maximum sorption capacity and drug release rate compared to CS-OA. The encapsulation efficiency of MTX from CS-CA was also higher than that of CS-OA.

Poly (diglycerol adipate) variants as enhanced nanocarrier replacements in drug delivery applications

Summary: Sustainably derived poly(glycerol adipate) (PGA) possesses desirable features for drug delivery, such as biodegradability, biocompatibility, self-assembly into nanoparticles, and functionalizable pendant groups. However, PGA suffers from poor amphiphilic balance, resulting in weak drug-polymer interactions, low drug-loading in nanoparticles, and low stability. To overcome these drawbacks, the polyester backbone was significantly modified while maintaining mild and sustainable polymerization conditions. Novel poly(diglycerol adipate) (PDGA) variants with hydrophilic diglycerol and hydrophobic hexanediol showed improved water solubility, diminished self-assembling ability, and enhanced features as nanocarriers. The PDGAHex nanoparticles exhibited stability in different environments and enhanced drug loading, as well as good biocompatibility in vitro and in vivo experiments.

Delivery of acetogenin-enriched Annona muricata Linn leaf extract by folic acid-conjugated and triphenylphosphonium-conjugated poly(glycerol adipate) nanoparticles to enhance toxicity against ovarian cancer cells

Summary: The study demonstrated the fabrication of poly(glycerol adipate) nanoparticles decorated with folic acid and triphenylphosphonium and their potential for delivering acetogenin-enriched leaf extract to ovarian cancer cells. The nanoparticles exhibited small size, narrow size distribution, high negative surface charge, and a biphasic sustained release of the extract. The FOL/TPP-decorated nanoparticles showed higher drug loading, enhanced toxicity to cancer cells, and improved cellular uptake compared to FOL-decorated nanoparticles. The FOL/TPP-NPs also demonstrated better stability, uptake efficiency, and targeting ability to mitochondria.

Summary: Poly(lactic-co-glycolic acid) (PLGA) is a commonly used polymer in drug delivery systems, exhibiting excellent biocompatibility, biodegradability, and spatio-temporal control of drug release. However, there is no general model that can describe all types of drug release in polymeric drug delivery systems. This study compared several mathematical models and found that the Hyperbolic Tangent Function model best fitted the drug release profile of PLGA particles synthesized by nanoprecipitation method.

A straightforward one-pot synthetic modification using a small library of diols along with glycerol was designed to alter the backbone of the polymer without affecting the hydrophilic glyceryl portion. Introducing additional functionality along the polymer backbone can alter the reactivity of the final polymer and the properties of the nanoparticles.
