Protein glycosylation alterations in hepatocellular carcinoma: function and clinical implications

Summary: Hepatocellular carcinoma (HCC) is a major cause of cancer-related deaths globally. Protein glycosylation, a common post-translational modification, plays a crucial role in regulating various biological processes and is implicated in the development of HCC. Aberrant protein glycosylation in hepatocytes contributes to the malignant transformation to HCC by modulating tumor-promoting signaling pathways. Changes in protein glycosylation could serve as potential diagnostic, prognostic, and therapeutic factors in HCC.

Summary: Aberrant changes in site-specific core fucosylation (CF) of serum proteins can serve as potential diagnostic markers of tumors. A study employed an optimized data-dependent acquisition (DDA) workflow to identify CF of serum proteins in patients with nonalcoholic steatohepatitis-related cirrhosis and hepatocellular carcinoma. The study found significant changes in CF peptides between cirrhosis and HCC, with one peptide showing the best diagnostic performance in discriminating HCC from cirrhosis.

A Fucosylated Glycopeptide as a Candidate Biomarker for Early Diagnosis of NASH Hepatocellular Carcinoma Using a Stepped HCD Method and PRM Evaluation

Summary: Aberrant specific N-glycosylation, particularly the increase in fucosylation on specific peptide sites of serum proteins, has been investigated as potential markers for diagnosing nonalcoholic steatohepatitis (NASH)-related hepatocellular carcinoma (HCC). In this study, a workflow combining broad scale marker discovery in serum and targeted marker evaluation was utilized to identify N-glycopeptides and determine N-glycosylation changes between disease states. The fucosylation level of the glycoprotein ceruloplasmin was evaluated and a specific tri-antennary glycopeptide showed a significantly increased fucosylation ratio in early HCC compared to cirrhosis. This tri-antennary glycopeptide in ceruloplasmin could serve as a potential biomarker for early detection of NASH HCC.

Summary: Nonalcoholic steatohepatitis (NASH) is the fastest growing cause of hepatocellular carcinoma (HCC) in the United States. Changes in N-glycosylation on specific glycosites of serum proteins have been investigated as potential markers for the early detection of NASH-related HCC. A glycopeptide with a Sialyl Lewis structure derived from serum haptoglobin (Hp) has been identified as a potential marker for NASH related HCCs. The combination of N241_A4G4F2S4 and alpha-fetoprotein (AFP) showed improved performance for HCC detection compared to AFP alone.
