Local complement synthesis-A process with near and far consequences for ischemia reperfusion injury and transplantation

Summary: For many years, the broader influence of complement in organ transplantation has been concealed, with a focus on its role as a target for circulating complement at the site of injury. However, recent studies on locally synthesized complement have shed new light on complement's participation in various aspects of transplantation. The precise complement activation pathways involved, their triggers, and their relevance still lack clarity. This review will present evidence for the emerging role of the lectin pathway, alongside the established role of the alternative pathway, and discuss the debate on precisely targeted versus broadly acting therapeutic innovation in transplantation.

Fucose as a new therapeutic target in renal transplantation

Summary: Ischaemia/reperfusion injury during kidney transplantation leads to delayed graft function and increased risk of graft loss. The activation of the complement system, particularly through CL-11 binding to a fucosylated ligand on renal tubules, contributes to tubular damage and reduced kidney function. Increasing free fucose in the kidney can act as a decoy molecule, reducing the clinical consequences of IRI mediated by CL-11.

Role of collectin-11 in innate defence against uropathogenic Escherichia coli infection

Summary: Classical collectins, including surfactant protein A and D, play a significant role in innate immunity and host defence against uropathogenic Escherichia coli-induced urinary tract infections (UTI). However, the role of collectin-11 (CL-11) in UPEC and UTI remains largely unexplored. This study demonstrates that CL-11 functions as a mediator of innate immunity, directly impacting bacterial growth and invasion while also modulating inflammatory responses during UTI. Additionally, CL-11 appears to regulate inflammatory cytokine production through pathways such as p38 MAPK phosphorylation, providing a potential target for UTI defense mechanisms.
