Congenital aniridia beyond black eyes: From phenotype and novel genetic mechanisms to innovative therapeutic approaches

Summary: Congenital PAX6-aniridia is not only characterized by the absence of the iris, but it is also associated with other ocular abnormalities and systemic features, making it a complex syndromic disorder. Foveal hypoplasia is now recognized as a more frequent feature than iris hypoplasia and plays a major role in visual prognosis. Genetic diagnosis of aniridia is challenging due to the genetic complexity caused by PAX6-related developmental genes. Targeted Next-Generation Sequencing combined with Whole-Genome Sequencing can increase the genetic diagnostic yield of aniridia.

Summary: This study compares the influence of airway epithelial cell secretions from COPD patients and non-COPD patients on macrophage phagocytic activity, and identifies the WNT/beta-catenin signalling-modulated and epithelium-derived S100A8/A9 as a potent regulator of macrophage phenotype and function. The findings suggest that COPD patients have a reduced capacity to support the phagocytic function of macrophages in response to acute bacterial exposure.

The product showed high percentages of cells expressing critical transcription factors for LSC stemness maintenance and corneal epithelial differentiation. Preclinical studies confirmed the engraftment potential of the cells and demonstrated their safety, leading to approval for a clinical trial to evaluate safety and therapeutic efficacy in patients with LSCD.
