Sayali S. Onkar et al.

Summary: Breast cancer is a diverse disease with different subtypes, and recent advances have provided new insights into the complex interactions between tumor and immune cells. While immunotherapy has shown limited promise for breast cancer, this review aims to identify opportunities for tailored immunotherapy agents based on the specific features of each breast cancer subtype.

Alcohol and breast cancer

Beata Starek-Swiechowicz et al.

Summary: Breast cancer is a leading cause of death in women worldwide, and alcohol plays a significant role in the development of tumors. This paper discusses the metabolism of alcohol and the mechanisms behind mammary tumorigenesis caused by alcohol. It presents numerous signaling pathways involved in neoplastic transformation following alcohol consumption in women and describes primary and secondary prevention, phytochemicals, synthetic chemicals, specific enzyme inhibitors, and selective receptor modulators.

Non-alcoholic fatty liver disease and increased risk of incident extrahepatic cancers: a meta-analysis of observational cohort studies

Alessandro Mantovani et al.

Summary: This meta-analysis found that NAFLD was significantly associated with an increased risk of developing gastrointestinal cancers (esophagus, stomach, pancreas, or colorectal cancers) by nearly 1.5-fold to 2-fold, as well as an increased risk of developing lung, breast, gynecological, or urinary system cancers by approximately 1.2-fold to 1.5-fold. These risks were independent of age, sex, smoking, obesity, diabetes, or other potential confounders.

Non-alcoholic fatty liver disease (NAFLD): a review of pathophysiology, clinical management and effects of weight loss

Sjaak Pouwels et al.

Summary: Given the increasing prevalence of diabetes and obesity worldwide, the deleterious effects of non-alcoholic fatty liver disease (NAFLD) are becoming a growing challenge for public health. NAFLD is closely associated with metabolic disorders and can range from mild forms to severe forms with potential liver failure. Advanced fibrosis is a major predictor of morbidity and liver-related mortality. Diagnosis of NAFLD is established through liver biopsy and treatment options include weight loss, dietary modification, and addressing underlying metabolic syndrome.

Non-alcoholic fatty liver disease through the female lifespan: the role of sex hormones

K. Pafili et al.

Summary: The prevalence of non-alcoholic fatty liver disease (NAFLD) varies among different stages of the female lifespan. This review summarizes the current evidence on the association between NAFLD and circulating sex hormones and explores the pathogenesis of NAFLD in relation to hormonal changes during reproductive, post-reproductive, and beyond. It also examines the effects of hormone therapies on NAFLD in women on menopausal hormone treatment or endocrine therapy after breast cancer. The findings suggest that fluctuations in estrogen levels, relative androgen excess, and age-related reduction in sex hormone-binding globulin are associated with increased NAFLD risk. Additionally, changes in body composition and insulin resistance during the peri-menopausal period may contribute to the increased risk of NAFLD. Further research is needed to identify subgroups at highest risk for NAFLD development and progression and to determine the role of hormone therapies, such as menopausal hormone treatment.

Hepatocellular and extrahepatic cancers in non-alcoholic fatty liver disease: A systematic review and meta-analysis

James A. Thomas et al.

Summary: Non-alcoholic fatty liver disease (NAFLD) is associated with hepatocellular carcinoma (HCC) and extrahepatic cancer. The incidence rate of HCC is higher in patients with advanced liver fibrosis or cirrhosis, while the incidence rate of extrahepatic cancers is even higher. These findings emphasize the importance of prevention and early detection of cancer in adults with NAFLD.

Summary: Nonalcoholic fatty liver disease (NAFLD) is the most prevalent chronic liver disease worldwide and is closely associated with insulin resistance and diabetes. Preventing ovariectomy-induced weight gain decreases tumor burden in rodent models of obesity and postmenopausal breast cancer

Elizabeth A. Wellberg et al.

Summary: Preventing weight gain after ovariectomy can decrease the development and growth of obesity-associated breast tumors, reduce visceral fat accumulation and insulin resistance. It also decreases the levels of tumor-promoting growth factors and inflammatory cytokines, and reduces the expression of tumor-promoting factors in mammary adipose tissue.

Non-alcoholic fatty liver disease and risk of fatal and non-fatal cardiovascular events: an updated systematic review and meta-analysis

Alessandro Mantovani et al.

Summary: Studies have shown a significant association between non-alcoholic fatty liver disease (NAFLD) and increased risk of cardiovascular disease (CVD) incidence. The risk of fatal or non-fatal CVD events is moderately increased with NAFLD, and this risk escalates with the severity of NAFLD, particularly with higher fibrosis stages. These findings suggest that NAFLD may be an independent risk factor for CVD morbidity and mortality.

PGC-1α activates SIRT3 to modulate cell proliferation and glycolytic metabolism in breast cancer

Yan Zu et al.

Summary: In this study, lower expression levels of PGC-1 alpha and SIRT3 were observed in breast cancer patients. Overexpression of PGC-1 alpha or SIRT3 significantly promoted apoptosis and inhibited proliferation in breast cancer cells. Furthermore, both PGC-1 alpha and SIRT3 were found to induce inhibition of glycolysis metabolism, with SIRT3 expression positively regulated by PGC-1 alpha.
