Cancer-associated fibroblast-dependent and -independent invasion of gastric cancer cells

Ryotaro Kondo et al.

Summary: This study investigated the invasive modes of gastric cancer cells in the presence of cancer-associated fibroblasts (CAFs), and found that gastric cancer cells exhibit both CAF-dependent and -independent modes of invasion, which are determined by intrinsic factors of the cancer cells. By modifying the invasive capacity of CAFs, the invasive mode of gastric cancer cells can be altered.

Systemic sclerosis

Elizabeth R. Volkmann et al.

Summary: Systemic sclerosis, also known as scleroderma, is a rare and complex autoimmune connective-tissue disease. Research advancements have improved our understanding of its disease pathogenesis and clinical phenotypes and expanded our treatment options. Early and accurate diagnosis, as well as ongoing risk stratification, play a crucial role in predicting organ involvement and disease progression. Treatment should be tailored to individual patients, considering their goals of care, and balancing side effects with efficacy.

Endothelial-to-mesenchymal transition in tumour progression and its potential roles in tumour therapy

Zeli Yin et al.

Summary: Tumour-associated endothelial cells (TECs) are a critical stromal cell type that play important roles in tumour angiogenesis. TECs have the ability to undergo endothelial-to-mesenchymal transition (EndoMT), which influences various malignant properties of tumours, making it a valuable target for tumour treatment. This review comprehensively explores the phenomenon of EndoMT in the tumour microenvironment, identifies the factors and mechanisms responsible for EndoMT induction, and discusses potential therapeutic strategies for targeting EndoMT in tumour treatment.

Podoplanin promotes the carcinogenicity of gastric cancer by activating ezrin and mediating the crosstalk between tumour cells and cancer-associated fibroblasts

Yueli Tian et al.

Summary: The role and biological mechanism of PDPN in the progression of gastric cancer were revealed in this study. PDPN acted as a promoter in the progression of gastric cancer through the activation of Ezrin expression and CAFs, providing a new route for gene-targeted therapy. These findings have important implications for the treatment of gastric cancer.

Differential regulation of lymphatic junctional morphology and the potential effects on cardiovascular diseases

Amanda M. Peluzzo et al.

Summary: The lymphatic vasculature is responsible for draining fluid, macromolecules, and immune cells from the interstitium as lymph and returning it to the bloodstream. This process is facilitated by a complex network of lymphatic vessels with differential regulation of cell-cell junctions. Different sections of the lymphatic bed have varying levels of permeability, controlled in part by their junctional morphology. Understanding the regulation of lymphatic junctional morphology is important for studying lymphatic permeability during development and disease, as well as its potential impact on cardiovascular diseases such as atherosclerosis.

From lymphatic endothelial cell migration to formation of tubular lymphatic vascular network

Tomohiro Shiiya et al.

Summary: During development, lymphatic endothelial cell (LEC) progenitors differentiate from venous endothelial cells only in limited regions of the body. LEC migration and subsequent tube formation are essential processes for the development of tubular lymphatic vascular network throughout the body. This review discusses the chemotactic factors, LEC-extracellular matrix interactions, and planar cell polarity regulating LEC migration and formation of tubular lymphatic vessels, providing insights into the molecular mechanisms underlying physiological lymphatic vascular development and lymphangiogenesis associated with pathological conditions.

Hypoxia-Inducible Factor-1a (HIF-1a) as a Biomarker for Changes in Microcirculation in Individuals with Systemic Sclerosis

Magdalena Maciejewska et al.

Summary: This study investigated the potential abnormalities of HIF-1a plasma concentration and its possible association with disease activity and vascular abnormalities in patients with systemic sclerosis. The results showed a marked increase in HIF-1a levels in patients with systemic sclerosis, especially in those with active disease and no history of digital ulcers. These findings suggest that HIF-1a may serve as a biomarker for assessing microcirculatory changes in individuals with systemic sclerosis.

Single-Cell Analysis of ADSC Interactions with Fibroblasts and Endothelial Cells in Scleroderma Skin

Marvin L. Frommer et al.

Summary: Adipose-derived stem cells (ADSCs) have anti-fibrotic and anti-inflammatory effects, but the mechanisms of action are not well understood. In this study, interactions between ADSCs, fibroblasts, and endothelial cells (EC) from scleroderma (SSc) skin were simulated in silico to investigate these mechanisms. Single-cell RNA sequencing data from patients with SSc and controls were analyzed, and differentially expressed genes and functional enrichment were identified. The study revealed pro-inflammatory and pro-fibrotic transcriptomes of fibroblasts and ECs in SSc, and highlighted the role of ECM proteins in interactions between ADSCs and these cell types. The findings suggest that the secretome of ADSCs may disrupt inflammatory and fibrotic processes in SSc by promoting angiogenesis and lymphangiogenesis.

Lymphatic Endothelial Markers and Tumor Lymphangiogenesis Assessment in Human Breast Cancer

Jia-Mei Chen et al.

Summary: Metastasis via lymphatic or blood vessels is the leading cause of death for breast cancer. However, the research progress for tumor lymphangiogenesis has lagged behind that for angiogenesis due to the lack of specific markers. With the discovery of lymphatic endothelial cell (LEC) markers, it has been shown that LEC plays an active role in various processes related to lymphatic formation, tumor growth, invasion, and antitumor immunity.

New Insights into Profibrotic Myofibroblast Formation in Systemic Sclerosis: When the Vascular Wall Becomes the Enemy

Eloisa Romano et al.

Summary: In systemic sclerosis (SSc), abnormalities in microvessel morphology evolve into a distinctive vasculopathy that progresses alongside tissue fibrosis orchestrated by myofibroblasts. The endothelial-to-mesenchymal transition (EndoMT) process may play a central role in the pathogenesis of SSc, contributing to both fibrotic vascular lesions and tissue fibrosis. A deeper understanding of the mechanisms underlying myofibroblast differentiation inside vessel walls provides the basis for targeted therapeutic strategies for SSc.

Role of Podoplanin-Positive Cells in Cardiac Fibrosis and Angiogenesis After Ischemia

Maria Cimini et al.

Summary: New insights into the cellular and extra-cellular composition of scar tissue after myocardial infarction have identified a heterogeneous podoplanin-expressing cell population associated with fibrogenic and inflammatory responses during scar formation. Podoplanin, mainly expressed by cardiac lymphatic endothelial cells in adult mouse hearts, is unexpectedly acquired by different types of cells after MI, suggesting an activation of progenitor cells during post-ischemic myocardial wound repair. Inhibition of the interaction between podoplanin-expressing cells and podoplanin-binding cells using podoplanin-neutralizing antibodies showed reduced inflammation post-MI and improved heart function and scar composition.
