p62/SQSTM1 in liver diseases: the usual suspect with multifarious identities

Chong Teik Tan et al.

Summary: Sequestosome-1 (SQSTM1), also known as p62, is an autophagy receptor that plays a crucial role in the pathogenesis of liver diseases such as NASH and HCC. Abundance of p62 inclusion bodies has been recognized as a biomarker for these diseases. However, the underlying mechanisms by which p62 regulates and contributes to the development and progression of these diseases remain incompletely understood. This review critically summarizes the findings from preclinical models and explores the potential of p62 as a predictive biomarker and therapeutic target for liver diseases.

Hepatocellular carcinoma and hepatitis C virus treatments: The bold and the beautiful

Walaa Abdelhamed et al.

Summary: Hepatocellular carcinoma (HCC) is a serious complication of hepatitis C virus (HCV) infection. Effective antiviral medications have made sustained viral response (SVR) or cure a realistic therapeutic goal for most chronic HCV patients. Direct-acting antivirals (DAAs) have changed the treatment landscape, reaching high rates of SVR with few side effects. The likelihood of HCC recurrence after HCV eradication appears to decrease over time following SVR. However, there is a need for markers/scores to predict the long-term risk of HCC in patients with HCV-related liver disease who attain SVR with DAAs.

Summary: The mechanisms of high fat diet (HFD)-induced non-alcoholic fatty liver disease (NAFLD) involve autophagy inhibition, NRF2 suppression, and promotion of ferroptosis. Activation of NRF2 can counteract the NRF2 suppression and ferroptotic cell death caused by HFD. The regulation of NRF2 by saturated fatty acids (SFAs) versus monounsaturated fatty acids (MUFAs) may explain their different effects.

Aging plays a crucial role in the development and progression of NAFLD, HCC, and other age-related liver diseases. The involvement of senescent cells and aging-related factors, such as NAD+, sirtuins, and mTOR, in the development of NAFLD has been explored, although their significance remains underestimated. This review summarizes current research on the roles of aging, cellular senescence, and aging-related factors in the evolution of NAFLD to HCC, with the aim of identifying potential therapeutic targets for aging-related NAFLD and its progression.

Transcription factor EB inhibits non-alcoholic fatty liver disease through fibroblast growth factor 21

Qi Gong et al.

Summary: We explored the potential role of transcription factor EB (TFEB) in the pathogenesis of non-alcoholic fatty liver disease (NAFLD), and found that fibroblast growth factor 21 (FGF21) has a significantly positive effect on NAFLD. Starvation led to an increase in liver lipids, which was reversed by re-feeding. Phosphorylated mTOR, ribosomal S6 kinase, TFEB, and FGF21 are involved in this process. The increased expression of TFEB in NAFLD mice by tail vein injection of Ad-TFEB could reverse lipid deposition and metabolic changes in NAFLD mice. TFEB upregulated FGF21 expression by targeting the promoter of FGF21. This study adds to our understanding of the potential role of TFEB in the progression of NAFLD, and may lay a basis for identifying new drug targets for NAFLD treatment.

Summary: This study investigated the inhibitory effect of red pepper seed extract (RPS) on hepatic lipid accumulation. The results showed that RPS treatment suppressed lipogenesis and induced hepatic autophagy, leading to a reduction in lipid accumulation. This effect was mediated through the AMPK-dependent pathway. In vitro experiments also demonstrated the ability of RPS to decrease lipid accumulation and downregulate lipogenesis-associated proteins in an oleic acid-induced hepatic steatosis cell model.

Empagliflozin activates Sestrin2-mediated AMPK/mTOR pathway and ameliorates lipid accumulation in obesity-related nonalcoholic fatty liver disease

Yuting Ma et al.

Summary: Empagliflozin (EMPA) therapy has been shown to improve non-alcoholic fatty liver disease (NAFLD) patients. This study investigated the functional implications of EMPA on the pathogenesis of NAFLD and identified the underlying molecular mechanisms. The results showed that EMPA treatment reversed liver damage by upregulating Sestrin2 and activating the AMPK-mTOR pathway, leading to the inhibition of lipogenesis and inflammation. These findings suggest that EMPA therapy could target Sestrin2 to alleviate lipogenesis and inflammation in obesity-related NAFLD.

Follistatin Alleviates Hepatic Steatosis in NAFLD via the mTOR Dependent Pathway

Junlu Tong et al.

Summary: The study revealed that hepatic FST expression is increased in NAFLD patients compared to normal samples. FST can alleviate hepatic steatosis by regulating the Akt/mTOR signaling pathway, suggesting that gene therapy for FST may be a promising treatment option for NAFLD.

Long non-coding RNA SNHG6 couples cholesterol sensing with mTORC1 activation in hepatocellular carcinoma

Fangzhou Liu et al.

Summary: Alterations in cholesterol homeostasis may contribute to hepatic cancer aggressiveness. The long non-coding RNA SNHG6 is found to be an important player in this process, linking cholesterol sensing with cancer cell growth through mTORC1 signaling. SNHG6 modulates cholesterol-induced mTORC1 activation by forming a SNHG6-FAF2-mTOR complex, enhancing cholesterol-dependent mTORC1 lysosomal recruitment and activation. Loss of SNHG6 inhibits mTORC1 signaling and impairs liver cancer tumor growth, suggesting SNHG6 as a potential target for liver cancer treatment.

Aberrant cholesterol metabolic signaling impairs antitumor immunosurveillance through natural killer T cell dysfunction in obese liver

Wenshu Tang et al.

Summary: Obesity and hypercholesterolemia are major risk factors for hepatocellular carcinoma. This study found that obesity leads to hepatic cholesterol accumulation, which suppresses natural killer T (NKT) cell-mediated antitumor immunosurveillance. By lowering cholesterol and inhibiting cholesterol biosynthesis, the function of NKT cells can be restored to prevent HCC development promoted by obesity.

Cache Domain Containing 1 Is a Novel Marker of Non-Alcoholic Steatohepatitis-Associated Hepatocarcinogenesis

Anna Kakehashi et al.

Summary: In this study, novel early molecular biomarkers of liver neoplasms in non-alcoholic steatohepatitis (NASH) were discovered using the STAM mouse model. The protein CACHD1 was found to be significantly elevated in hepatocellular carcinomas, suggesting its potential as a biomarker and molecular target in NASH-associated hepatocarcinogenesis involving cell proliferation, autophagy, and apoptosis. The research highlights CACHD1 as an early biomarker of liver preneoplastic and neoplastic lesions in the context of NASH, with implications for potential therapeutic targets in diabetes mellitus/NASH-associated hepatocarcinogenesis.

Loss of hepatic Flcn protects against fibrosis and inflammation by activating autophagy pathways

Mathieu Paquette et al.

Summary: FLCN protein activity could be a promising target for small molecule drugs to treat liver fibrosis by specifically activating autophagy, promoting the degradation of intracellular lipid stores, ultimately resulting in reduced hepatocellular damage and inflammation. These results show an unexpected role for Flcn in fatty liver disease progression and highlight new potential treatment strategies.
