Target therapy for BRAF mutated anaplastic thyroid cancer: a clinical and molecular study

Tiago Nunes da Silva et al.

Summary: This study aimed to evaluate the efficacy of Dabrafenib plus Trametinib in patients with BRAF-positive anaplastic thyroid carcinoma (ATC) and compare the outcomes with standard treatment. The results showed that patients in the Dabrafenib plus Trametinib group had better overall survival and progression-free survival compared to standard treatment, with a good safety profile.

Translating the immune microenvironment of thyroid cancer into clinical practice

Lucas Leite Cunha et al.

Summary: Thyroid cancer provides a valuable model for studying tumor immune microenvironment due to its immune response characteristics. The cellular composition of the microenvironment is associated with the clinical aggressiveness of the cancer, and genetic signatures can be used to develop prognostic models. Immunotherapy shows promise as a treatment strategy, especially for advanced thyroid cancer, but further research is needed to improve its effectiveness.

Macrophage-Based Approaches for Cancer Immunotherapy

Nicholas R. Anderson et al.

Summary: Adoptive cell therapy with genetically modified T cells has shown promising results in hematologic malignancies, but applying it to solid tumors has been challenging. This has led to the investigation of alternative immune cells as therapeutic options. Macrophages have functional plasticity that can be harnessed for both antitumor and protumor effects, leading to efforts to deplete or repolarize tumor-associated macrophages, as well as exploring the potential of adoptively transferring genetically modified macrophages.

Serum Follistatin Is Increased in Thyroid Cancer and Is Associated With Adverse Tumor Characteristics in Humans

Grigorios Panagiotou et al.

Summary: Obesity and classical growth factors are associated with thyroid cancer, while novel hormones such as follistatins and activins may play a role. This study found that serum levels of follistatin were increased in thyroid cancer patients and correlated with tumor aggressiveness, including vascular invasion and metastases. Further longitudinal studies are needed to confirm these findings.

Anaplastic Thyroid Carcinoma: Current Issues in Genomics and Therapeutics

Ichiro Abe et al.

Summary: Anaplastic thyroid carcinoma is a highly aggressive type of thyroid cancer, with common mutations in genes such as p53 and TERT. Current treatment options include curative resection, radiotherapy, and combination chemotherapies, with ongoing research on molecular targeted therapies and immunotherapy showing promise in managing this challenging cancer.

Secreted Factors by Anaplastic Thyroid Cancer Cells Induce Tumor-Promoting M2-like Macrophage Polarization through a TIM3-Dependent Mechanism

Cinthia Carolina Stempin et al.

Summary: Anaplastic thyroid cancer (ATC) is a highly aggressive type of thyroid cancer, and there is a critical need for new therapeutic targets. Tumor-associated macrophages (TAMs) are heavily infiltrated in ATCs and may play a role in promoting tumor progression, with TIM3 identified as a potential novel target for treating ATC.

Identification of SPRY4 as a Novel Candidate Susceptibility Gene for Familial Nonmedullary Thyroid Cancer

Ines J. Marques et al.

Summary: Through whole-exome sequencing analysis, SPRY4 gene was identified as a potentially novel candidate susceptibility gene for familial nonmedullary thyroid cancer. In vitro experiments indicated that the SPRY4 variant resulted in increased cell viability and clonogenic capacity, mediated through the mitogen-activated protein kinase/extracellular signal-regulated kinase pathway.

Immune Landscape of Thyroid Cancers: New Insights

Elisa Menicali et al.

Summary: The immune system plays a crucial role in cancer prevention, initiation, and progression, with thyroid tumor cells utilizing various mechanisms to evade immune destruction. Studies have shown the recruitment of immunosuppressive cells and the expression of negative immune checkpoints in the thyroid cancer microenvironment. Understanding the interactions between tumor and microenvironment is crucial for directing immunotherapeutic approaches in treating thyroid cancer.

Regulation of Cancer Metastasis by TRAIL/Death Receptor Signaling

You-Take Oh et al.

Summary: The involvement of the TRAIL/death receptor signaling pathway in the regulation of cancer invasion and metastasis is complex, with both positive and negative roles reported. The underlying molecular mechanisms are even more complicated. This review focuses on discussing the current understanding of how TRAIL/death receptor-mediated signaling regulates cancer cell invasion and metastasis.

M2-like tumour-associated macrophage-secreted IGF promotes thyroid cancer stemness and metastasis by activating the PI3K/AKT/mTOR pathway

Juan Lv et al.

Summary: The study revealed that M2-like tumour-associated macrophages (TAMs) can accelerate metastasis and increase stemness of anaplastic thyroid carcinoma (ATC) cells, potentially through the secretion of IGF that activates the IR-A/IGF1R-mediated PI3K/AKT/mTOR signalling pathway.
