Loss of MAT2A compromises methionine metabolism and represents a vulnerability in H3K27M mutant glioma by modulating the epigenome

Brian J. Golbourn et al.

Summary: Agnihotri and colleagues demonstrate that the loss of the MAT2A enzyme in the methionine cycle leads to a depletion of H3K36me3 and increases survival in glioma models. They identify MAT2A as a critical vulnerability in H3K27M gliomas through an RNA screen and show that the depletion of MAT2A reduces H3K36me3 levels, impacting oncogenic and developmental transcription programs. Methionine-restricted diets also extend survival in DMG models.

BATF3 promotes malignant phenotype of colorectal cancer through the S1PR1/p-STAT3/miR-155-3p/WDR82 axis

Ping Li et al.

Summary: The study elucidated the oncogenic role of BATF3 in CRC by activating the S1PR1/p-STAT3/miR-155-3p/WDR82 axis, promoting the proliferation, migration, and invasion of CRC cells. In vivo assays further validated the in vitro findings, indicating that inactivation of the BATF3/S1PR1/p-STAT3/miR-155-3p/WDR82 axis suppresses CRC tumor growth.

M2 macrophages-derived exosomal microRNA-501-3p promotes the progression of lung cancer via targeting WD repeat domain 82

Jie Lei et al.

Summary: Research suggests that exosomal miR-501-3p derived from M2 macrophages promotes the progression of lung cancer (LC) by downregulating WDR82. This study provides important insights into the molecular mechanisms underlying the development of LC.

Epigenetically defined therapeutic targeting in H3.3G34R/V high-grade gliomas

Stefan R. Sweha et al.

Summary: High-grade gliomas with H3.3G34R/V mutations have poor prognosis and current treatments are not curative. Epigenetic analysis revealed the LIF/STAT3 pathway as a key vulnerability in H3.3G34R/V gliomas, suggesting potential for targeted therapies.

ADAR1 is a new target of METTL3 and plays a pro-oncogenic role in glioblastoma by an editing-independent mechanism

Valentina Tassinari et al.

Summary: The study reveals that METTL3 methylates ADAR1 mRNA and increases its protein level, linking m6A and A-to-I editing and promoting glioblastoma growth. ADAR1 plays a crucial role in cancer progression and its knockdown can strongly inhibit glioblastoma growth.
