Systematic analysis of factors that improve homologous direct repair (HDR) efficiency in CRISPR/Cas9 technique

Summary: The CRISPR/Cas9 bacterial system is a powerful tool for genetic manipulation, but HDR efficiency can be lower than random indel creation. This study demonstrates the synergistic effects of using double sgRNA, cell synchronization cycle, and rational design of ssODN to enhance HDR efficiency, particularly in editing the TNF alpha gene. The use of two sgRNA with asymmetric donor design and triple transfection events significantly improved HDR efficiency, providing a new strategy for CRISPR/Cas9-mediated human genome editing.
