An Overview of Circulating Cell-Free Nucleic Acids in Diagnosis and Prognosis of Triple-Negative Breast Cancer

Domenico Tierno et al.

Summary: This review summarizes the potential of circulating tumor nucleic acids (ctNAs) analysis in aiding the diagnosis and prognosis of triple-negative breast cancer (TNBC). Mutations and copy number variations of TP53 and PIK3CA/AKT genes in plasma may serve as important markers for TNBC onset, progression, metastasis, and clinical follow-up. The expression profile of circulating tumor non-coding RNAs (ctncRNAs) can predict molecular subtypes of breast cancer and aid in the identification of TNBC.

How to conduct a systematic review and meta-analysis of prognostic model studies

Johanna A. A. Damen et al.

Summary: This article provides a step-by-step guidance for conducting and reading a systematic review of prognostic model studies. It also offers an overview of the methodology and guidance available for each step of the review process. A case study on prognostic models for COVID-19 patients is used to illustrate the process. The implications of these reviews for clinical practice and further research depend on the complete reporting of primary studies.

Prognostic and diagnostic values of non-coding RNAs as biomarkers for breast cancer: An umbrella review and pan-cancer analysis

Afshin Bahramy et al.

Summary: This study conducted an umbrella review on the prognostic and diagnostic value of non-coding RNAs (ncRNAs), specifically long non-coding RNAs (lncRNAs) and microRNAs (miRNAs), in breast cancer. The findings suggest that ncRNAs can serve as prognostic and diagnostic biomarkers for breast cancer patients, regardless of the sample sources, ethnicity of patients, and subtype of breast cancer.

The Role of miRNAs in the Prognosis of Triple-Negative Breast Cancer: A Systematic Review and Meta-Analysis

Talita Araujo B. da S. Santana et al.

Summary: Breast cancer is a common malignancy among women, and microRNAs (miRs) have been found to play a significant role in the prognosis of triple-negative breast cancer (TNBC). A systematic review of 43 articles with a total of 5421 TNBC patients revealed that low expression of miR-155 and high expression of miR-21 were associated with reduced overall survival, while high expression of miR-27a/b and miR-301a/b were associated with lower overall survival. Decreased expression of miR-30 and miR-374a/b were associated with worse relapse-free survival and shorter disease-free survival, respectively. These findings highlight the importance of microRNAs in the prognosis of TNBC.

Summary: Triple-negative breast cancer (TNBC) is an aggressive subtype with limited treatment options. Chemotherapy has been the mainstay, but recent studies have shown immune checkpoint inhibitors to be effective as first-line treatment for advanced TNBC expressing PD-L1. However, there are still many open questions regarding the use of immunotherapy in TNBC.

Potential Role of Circulating miRNAs for Breast Cancer Management in the Neoadjuvant Setting: A Road to Pave

Chiara Benvenuti et al.

Summary: In breast cancer management, neoadjuvant chemotherapy is widely used, but there is a lack of technology that can predict the benefits of neoadjuvant chemotherapy for breast cancer patients. Circulating miRNAs have emerged as potential non-invasive biomarkers for breast cancer and may be useful in diagnosing, predicting, and prognosticating breast cancer patients undergoing neoadjuvant chemotherapy. Among circulating miRNAs, miR-21-5p and miR-34a-5p are the most promising biomarkers for breast cancer patients undergoing neoadjuvant chemotherapy.

Comprehensive characterization of RNA cargo of extracellular vesicles in breast cancer patients undergoing neoadjuvant chemotherapy

Lilite Sadovska et al.

Summary: This study investigated the dynamics of extracellular vesicles (EVs) levels in the plasma of breast cancer (BC) patients undergoing neoadjuvant chemotherapy (NAC) and explored the relevance of their RNA cargo for predicting treatment response. The results showed that EV levels were associated with alterations in the proportions of various RNA biotypes in EVs. Several RNA biomarkers were identified for predicting response to NAC at the time of diagnosis, and potential RNA candidates were found for further study on their role in chemoresistance and metastasis.

Breast cancer cell-derived microRNA-155 suppresses tumor progression via enhancing immune cell recruitment and antitumor function

Junfeng Wang et al.

Summary: The expression of miR-155 in breast cancer cells is associated with a favorable antitumor immune profile and better patient outcomes. Overexpression of miR-155 in breast cancer cells enhances T cell influx, delays tumor growth, and increases sensitivity to immune checkpoint blockade therapy. The mechanism involves upregulation of CXCL9/10/11 production mediated by SOCS1 inhibition and increased phosphorylated STAT1/STAT3 ratios. Serum miR-155 levels in breast cancer patients correlate with tumor miR-155 levels and tumor immune status.
