Activated fibroblasts in cancer: Perspectives and challenges

Summary: Activated fibroblasts in tumors, known as cancer-associated fibroblasts (CAFs), have been a highly studied topic in the past decade. Additionally, the review highlights the need to consider CAF investigation as a promising therapeutic avenue alongside immune and cancer cells and outlines the challenges that need to be addressed for successful translation of new findings.

An Analysis of Nectin-4 (PVRL4) in Penile Squamous Cell Carcinoma

Landscape of surfaceome and endocytome in human glioma is divergent and depends on cellular spatial organization

Summary: Therapeutic strategies targeting tumor surfaceome (TS) have become promising approaches in cancer treatment. However, there is still a lack of comprehensive strategies to identify potential TS targets. In this study, we developed a platform called TS-MAP, which combined with a newly curated TS classifier called SURFME, to analyze primary 3D cultures and intact patient glioma tumors. TS-MAP could specifically identify proteins suitable for targeted therapies such as ADCs. The spatial organization of cells had a significant impact on the surfaceome and endocytome in high-grade gliomas, indicating the need for personalized approaches in target screening. This study provides valuable insights for the development of immunotherapy strategies and proteomics-based target identification.

Immune-based therapies in penile cancer

Head and neck cancer N-glycome traits are cell line and HPV status-dependent

Summary: Glycosylation, the most common post-translational modification of proteins, is closely associated with malignant epithelia such as head and neck squamous cell carcinoma (HNSCC). This study aimed to analyze the N-glycome of HNSCC cell lines and found that oligomannose-type N-glycans were the predominant features, while the levels of sialylated N-glycans varied among different cell lines. HPV-positive cell lines showed higher levels of phosphorylated oligomannose N-glycans, indicating an upregulation of lysosomal pathways. This systematic mapping of the N-glycome in HNSCC provides valuable insights for further diagnostic studies.
