Immune-related adverse effects of checkpoint immunotherapy and implications for the treatment of patients with cancer and autoimmune diseases

Betul Ibis et al.

Summary: In the past decade, antibody-based immunotherapies that modulate immune responses against tumors have revolutionized cancer treatment. However, these therapies have limitations due to their inability to selectively target the tumor microenvironment. Immune checkpoint inhibitors (ICI) have shown promising results in treating cancer, but their use has been limited by the occurrence of immune-related adverse effects (irAEs). Accumulating data suggests that ICI may be safely administered to patients with pre-existing autoimmune diseases (ADs). This review discusses the mechanisms of irAEs and the evolving knowledge of ICI therapy in patients with cancer and pre-existing ADs.

Metabolic modulation of immune checkpoints and novel therapeutic strategies in cancer

Yi Wang et al.

Summary: Cytotoxic T-lymphocyte-associated antigen-4 (CTLA-4) or programmed death-1 (PD-1)/programmed death-ligand 1 (PD-L1)-based immune checkpoint inhibitors (ICIs) have shown significant improvements in overall survival of certain cancer patients, but resistance to immunotherapy remains a challenge. This article examines the metabolic modulation of tumor immunity and immune checkpoints in the tumor microenvironment (TME) and discusses potential metabolism-based therapeutic strategies. Understanding the link between tumor metabolism and immunotherapy is crucial for cancer treatment and research.

Immune checkpoint modulators in cancer immunotherapy: recent advances and emerging concepts

Yuchen Wang et al.

Summary: The discovery of immune checkpoint inhibitors is considered a significant breakthrough in tumor therapy, but severe side effects and ineffective treatment outcomes still exist. In addition to known molecules like PD-1/PD-L1 and CTLA-4, novel immune checkpoint molecules such as VISTA and CD39 are being widely investigated for potential clinical benefits.

Interplay between Epstein-Barr virus infection and environmental xenobiotic exposure in cancer

Francisco Aguayo et al.

Summary: EBV, a herpesvirus associated with lymphoid and epithelial malignancies, requires additional factors for tumor development, with xenobiotics like tobacco smoke components being suggested as cofactors. This interaction provides insight into mechanisms of EBV involvement in carcinogenesis and potential prevention strategies.
