Changes of the subchondral bone microchannel network in early osteoarthritis

Summary: We investigated the 3D network of subchondral microchannels (CMMC) connecting the deep zone of cartilage to the bone marrow and its relevance to early idiopathic osteoarthritis (OA). Our quantitative analysis revealed that the number and morphology of CMMC were influenced by the region of the joint, while the size of CMMC was influenced by both the joint location and the cartilage condition. Moreover, we observed an enlargement of the microchannels in early OA, particularly in the non-load-bearing region and the peripheral rim of the femoral head.

Summary: Osteoarthritis is a joint disease characterized by the degradation of cartilage and loss of joint mobility. This study found that chondrocytes from osteoarthritic mice and patients express pro-inflammatory markers and RANKL in a manner dependent on NF-kappa B. Exposure of chondrocytes to IL-1 beta and bone particles, which mimic bone matrix breakdown products in synovial fluid, recapitulated the inflammatory phenotype of chondrocytes. Bone particles and IL-1 beta induced senescent and apoptotic changes in chondrocytes through oxidative stress from various cellular sources. These findings enhance our understanding of the inflammatory mechanisms involved in osteoarthritis.

Platelet-rich plasma injections delay the need for knee arthroplasty: a retrospective study and survival analysis
