The Role of l-Carnitine in Mitochondria, Prevention of Metabolic Inflexibility and Disease Initiation

Summary: Mitochondria play a critical role in cellular fate and metabolism, and their dysfunction is implicated in various diseases. l-carnitine, an important nutrient for proper mitochondrial function, is involved in fatty acid oxidation and has protective effects on cellular membranes. It also modulates ketogenesis, glucogenesis, and the elimination of toxic metabolites. Deficiency in l-carnitine is observed in many diseases and addressing nutrient deficiencies, like l-carnitine, holds promise for managing age and metabolic related diseases.

Inhibition of ferroptosis attenuates oligospermia in male Nrf2 knockout mice

Summary: Expression of Nrf2 in sperm is reduced in oligospermia patients, but the mechanism is unclear. This study found that Nrf2 and GPX4 protein expressions in sperm of oligospermia men were significantly decreased. Animal experiments showed that Nrf2 knockout in mice led to the occurrence of ferroptosis in spermatogenic cells, resulting in oligospermia. Treatment with a ferroptosis inhibitor reversed the decrease in sperm concentration and motility.

Relationship between functional Nrf2 gene promoter polymorphism and sperm DNA damage in male infertility

Summary: This study examines the association of the -617 C > A polymorphism in the Nrf2 gene with male infertility in a Turkish population. Results showed a significantly higher prevalence of the AA genotype in infertile men, especially in smokers, which was associated with reduced sperm motility and increased risk of oligoasthenozoospermia. Furthermore, smokers carrying the AA genotype were more likely to have high sperm-DNA damage, indicating the role of Nrf2 gene polymorphism as a risk factor for male infertility.
