STING inhibits the reactivation of dormant metastasis in lung adenocarcinoma

Jing Hu et al.

Summary: Metastasis often occurs when cancer cells that are dormant after primary tumor treatment become active again. The stimulator of interferon genes (STING) pathway has been identified as a suppressor of metastatic outbreak and a potential therapeutic target to prevent disease relapse. By understanding the determinants of immune reactivity during exit from dormancy, researchers have found that STING activity increases in metastatic progenitors that re-enter the cell cycle and can be dampened by methylation or chromatin repression. Activation of STING function in cancer cells derived from spontaneous metastases eliminates dormant metastasis and prevents spontaneous outbreaks.

Brief report: STING expressed in tumor and non-tumor compartments has distinct roles in regulating anti-tumor immunity

Jennie C. Kim et al.

Summary: STING pathway plays context-dependent roles in regulating anti-tumor immunity and response to immunotherapy. In poorly immunogenic tumor models, STING expression in non-tumor cells is crucial for productive anti-tumor immune responses. In immunogenic models, constitutive STING activation in tumor cells can partially bypass the requirement for STING-dependent activity from immune cells.

Leveraging STING, Batf3 Dendritic Cells, CXCR3 Ligands, and Other Components Related to Innate Immunity to Induce A Hot Tumor Microenvironment That Is Responsive to Immunotherapy

Robin Reschke et al.

Summary: The lack of T cells in tumors is one of the main reasons for the failure of immunotherapy. Strengthening innate immune mechanisms can improve the efficacy of tumor immunotherapy, such as delivering innate immune factors into the tumor microenvironment. These mechanisms include immune sensing in the tumor and efficient antigen presentation, as well as important cells and molecules that promote T cell inflamed tumor formation and immune therapy response.

cGAS-STING signaling encourages immune cell overcoming of fibroblast barricades in pancreatic cancer

Ayano Kabashima et al.

Summary: This study investigates the impact of cGAS-STING expression and the presence of cancer-associated fibroblasts (CAFs) on immune cell infiltration in pancreatic ductal adenocarcinoma (PDAC). The results show that PDAC cases with co-expression of cGAS and STING have favorable survival and exhibit cytotoxic CD8 + T cell infiltrations. However, the presence of CAFs impedes immune cell infiltration.

T cell-intrinsic STING signaling promotes regulatory T cell induction and immunosuppression by upregulating FOXP3 transcription in cervical cancer

Huanhe Ni et al.

Summary: In this study, it was found that increased STING and CCL22 levels, as well as higher levels of FOXP3(+) cells and lower levels of CD8(+) cells in tumor tissues of cervical cancer, predicted poor survival. Activation of STING signaling in cooperation with T cell receptor and other signals promoted iTreg differentiation in both human and murine CD4(+) naive T cells. Additionally, tumor-derived exosomes were found to activate STING signaling in tumor-infiltrated T cells, leading to iTreg expansion.

Summary: The activation of STING pathway promotes antitumor immunity, but STING agonists have not been successful in clinical trials. This study found that STING agonists can enhance the migration and killing ability of NK cells and CAR-NK cells in malignant pleural mesothelioma. It highlights the importance of using human tumor samples to evaluate immune therapies and supports combining STING agonists with other cellular therapies.

Autophagy, ferroptosis, pyroptosis, and necroptosis in tumor immunotherapy

Weitong Gao et al.

Summary: Immunotherapy has made significant breakthroughs in cancer treatment, but the poor response of some tumors limits its application. Inducing non-apoptotic regulated cell death (RCD) may convert these unresponsive tumors into those that respond to immunotherapy. Autophagy, ferroptosis, pyroptosis, and necroptosis have been found to have both inhibitory and synergistic effects on antitumor immune responses, suggesting that targeted therapies against these processes combined with immunotherapy may be effective even in resistant tumors.

Epigenetic reprogramming of tumor cell-intrinsic STING function sculpts antigenicity and T cell recognition of melanoma

Rana Falahat et al.

Summary: The lack or loss of tumor antigenicity is a key mechanism of immune evasion and resistance to T cell-based immunotherapies. Epigenetic regulation, specifically DNA methylation, plays a role in suppressing the STING signaling function in melanoma cells, but this suppression can be reversed through pharmacologic DNA methylation inhibition. Restoring STING signaling improves melanoma cell antigenicity and enhances recognition and killing by cytotoxic T cells, highlighting the functional significance of epigenetic processes in tumor-immune evasion.
