Commercial influenza vaccines vary in HA-complex structure and in induction of cross-reactive HA antibodies

Summary: Influenza viruses cause worldwide epidemics and yearly reformulation of commercial influenza vaccines (CIV) due to the antigenic variation of Hemagglutinin (HA). This study investigates the structural organization of HA in four current CIVs using electron microscopy, revealing different arrangements including individual HAs, starfish structures, and spiked-nanodisc structures. CIVs with spiked nanodiscs induce the highest levels of cross-reactive antibodies. Therefore, HA structural organization can be an important parameter for CIVs and their efficacy.

Summary: Seasonal influenza viruses cause 1 billion infections per year, with millions of severe cases and hundreds of thousands of deaths. Current influenza vaccines have varying effectiveness and target the HA and NA surface glycoproteins. This study developed a bioprocess to manufacture inactivated split cHA and mHA vaccines and a method to quantify HA with a prefusion stalk. The process demonstrated high yield and low levels of impurities, providing a basis for pre-clinical and future clinical trials.

Female-biased effects of aging on a chimeric hemagglutinin stalk-based universal influenza virus vaccine in mice

Summary: This study compares the effects of sex and age on universal influenza vaccine-induced immunity. The results show that adult mice develop more and higher quality antibodies compared to aged mice, providing better protection. Male mice experience less decline in immunity and protection compared to females. Transferring serum from young mice to aged mice cannot reverse this decline in immunity. Therefore, the sex-specific effects of aging on immunity are significant and should be considered in the development of universal vaccines.

The antibody titres were highest in the adjuvanted groups and higher after a two-dose schedule compared to a one-dose schedule.
