Rapid generation of Caenorhabditis elegans single-copy transgenes combining recombination-mediated cassette exchange and drug selection

Michael L. Nonet

Summary: This article presents a simplified method for inserting large, single-copy transgenes into the Caenorhabditis elegans genome using recombination-mediated cassette exchange (RMCE). It only relies on drug selection to obtain homozygous fluorescent protein (FP) marked transgenes in 3 generations with high efficiency. Landing sites on four chromosomes allow marking lines in different cell types. Various vectors and selection methods can be used to create transgenes expressing different colored FPs. The rapid RMCE toolbox provides a platform for developing further innovative uses of RMCE.

Impact of rapeseed pomace extract on markers of oxidative stress and DNA damage in human SH-SY5Y cells

Franziska Pohl et al.

Summary: The study highlights the neuroprotective properties of rapeseed pomace extract in human neuroblastoma cells, showing a potential for alleviating oxidative stress-induced cell damage. The extract was found to increase levels of certain protective proteins and downregulate stress-responsive proteins, indicating promising cell protective properties against oxidative stress.

Paraquat exposure over generation affects lifespan and reproduction through mitochondrial disruption in C. elegans

Snigdha Bora et al.

Summary: The study shows the effects of acute and chronic exposure to Paraquat on reproduction, longevity, gene expression, and mitochondrial physiology in C. elegans. Chronic exposure to Paraquat leads to a decline in fertility and a trade-off between reproduction and somatic maintenance. Mutations in CEP-1 and isp-1 influence the sensitivity and tolerance to chronic Paraquat exposure.

Mitochondrial hydrogen peroxide positively regulates neuropeptide secretion during diet-induced activation of the oxidative stress response

Qi Jia et al.

Summary: Mitochondria play a crucial role in connecting metabolism with neurotransmitter release. The study demonstrates that endogenous hydrogen peroxide produced by axonal mitochondria acts as a signaling molecule to selectively regulate neuropeptide secretion in C. elegans.
