Lung adenocarcinoma promotion by air pollutants

William Hill et al.

Summary: This study reveals a strong association between PM2.5 air pollutants and lung cancer risk, suggesting that these pollutants promote lung cancer development by acting on cells with pre-existing oncogenic mutations in healthy lung tissue. The mechanism involves the influx of macrophages and release of interleukin-1 beta, which induce a progenitor-like state in EGFR mutant lung alveolar type II epithelial cells, fueling tumorigenesis. The findings underscore the tumor-promoting role of PM2.5 air pollutants and emphasize the need for public health policy initiatives to address air pollution.

How to Increase Cellular Glutathione

Daniela Giustarini et al.

Summary: Glutathione (GSH) has special antioxidant properties due to its high concentration, ubiquity, and reactivity. In diseases where oxidative stress plays a role, GSH concentration is reduced, making cells more prone to damage. This review discusses strategies for increasing cellular GSH, including GSH itself, derivatives, activators, prodrugs, foods, and diets, as well as their mechanisms and advantages/disadvantages.

Targeting IL-1? as an immunopreventive and therapeutic modality for K-ras-mutant lung cancer

Bo Yuan et al.

Summary: The blockade of IL-1β has been found to significantly reduce lung tumor burden and reprogram the lung microenvironment in a mouse model of K-ras mutant lung adenocarcinoma, leading to increased infiltration of anti-tumor CD8+ T cells. In KM-LUAD patients, IL-1β expression is positively correlated with the infiltration of immunosuppressive cells and the expression of related genes.

Glutathionylation chemistry promotes interleukin-1 beta-mediated glycolytic reprogramming and pro-inflammatory signaling in lung epithelial cells

Reem Aboushousha et al.

Summary: The study reveals that IL1B induces glycolysis in airway epithelial cells, potentially affecting allergic airway inflammation and remodeling. Cells lacking the enzyme Glrx show modulation of multiple metabolic pathways and increased secretion of TSLP. Targeted inhibition of glycolysis prevents TSLP release, suggesting a potential therapeutic target in pulmonary pathologies.

Glutathione-S-transferase P promotes glycolysis in asthma in association with oxidation of pyruvate kinase M2

Cheryl van de Wetering et al.

Summary: The research demonstrates the role of GSTP in the pathogenesis of allergic airways disease by promoting glycolytic reprogramming and disrupting PKM2. Experimental results show that deletion or inhibition of GSTP can decrease secretion of pro-inflammatory mediators, reduce glycolytic flux, and potentially serve as a treatment target for allergic airways disease.

OPLS4: Improving Force Field Accuracy on Challenging Regimes of Chemical Space

Chao Lu et al.

Summary: The development and validation of the OPLS4 force field, which builds upon previous work with OPLS3e, improved model accuracy on challenging regimes of drug-like chemical space. A novel parametrization strategy for charged species was introduced, leading to improved accuracy on benchmarks assessing small-molecule solvation and protein-ligand binding.

GLS-driven glutamine catabolism contributes to prostate cancer radiosensitivity by regulating the redox state, stemness and ATG5-mediated autophagy

Anna Mukha et al.

Summary: Radiotherapy is an effective treatment for localized prostate cancer due to its ability to induce oxidative stress and DNA damage in tumor cells. However, resistance mechanisms and normal tissue toxicity can limit its effectiveness. Metabolic reprogramming plays a role in therapy resistance, with glutamine metabolism identified as a potential biomarker and therapeutic target for sensitizing prostate cancer to radiotherapy. Inhibiting glutamine catabolism can sensitize cancer cells while sparing nonmalignant cells, and targeting this pathway may also reduce cancer stem cell populations and enhance the effectiveness of radiation therapy.
