Astrocyte contribution to dysfunction, risk and progression in neurodegenerative disorders

Ashley N. Brandebura et al.

Summary: There is increasing evidence that astrocytes play a significant role in neurodegenerative disorders, both as initiators and contributors to disease progression. This review discusses the involvement of astrocytes in disorders such as Alzheimer's disease, Parkinson's disease, Huntington's disease, and amyotrophic lateral sclerosis, and explores their importance in supporting neuronal function in the healthy brain as well as the changes they undergo in pathological conditions.

Reactive astrocytes acquire neuroprotective as well as deleterious signatures in response to Tau and Aβ pathology

Zoeb Jiwaji et al.

Summary: Alzheimer's disease affects astrocytes, and both A beta and Tau pathology induce changes in astrocytes. A beta mainly influences the expression of AD risk genes, while both pathologies induce age-dependent changes and have overlapping features with astrocytes found in post-mortem AD patients. Both A beta and Tau pathology alter the characteristics of astrocytes, including repression of bioenergetic and translation machinery, induction of inflammation pathways, and expression of protein degradation/proteostasis genes. Astrocyte-specific expression of Nrf2, a protective factor, can recapitulate elements of these astrocyte changes and reduce A beta deposition and phospho-tau accumulation. This Nrf2 expression also improves brain-wide transcriptional deregulation, cellular pathology, neurodegeneration, and behavioral/cognitive deficits.

Lung-brain axis

Ousman Bajinka et al.

Summary: The appreciation of human microbiome is gaining strong grounds in biomedical research, focusing on the lung-brain axis in addition to the gut-brain axis. Interdisciplinary approaches based on emerging studies are recommended to develop effective treatments for lung induced neurodegenerative diseases. In-depth understanding of the bi-directional communication between host and microbiome in the lung could confer treatment to respiratory infections, alleviate stress, anxiety, and enhance neurological effects.

SARS-CoV-2 is associated with changes in brain structure in UK Biobank

Gwenaelle Douaud et al.

Summary: Through long-term observation of participants' MRI images, it was found that COVID-19 infection is associated with abnormal brain changes, including a reduction in gray matter thickness in the orbitofrontal cortex and parahippocampal gyrus, greater changes in markers of tissue damage in regions functionally connected to the primary olfactory cortex, and a greater reduction in global brain size. Infected individuals also showed significant cognitive decline.

Summary: This study analyzed the US Department of Veterans Affairs national healthcare databases to investigate the occurrence of Long COVID and death risk after breakthrough SARS-CoV-2 infection. The findings showed that vaccination significantly reduced the risk of death and post-acute sequelae but did not eliminate them completely. These results underscore the importance of further research on prevention and post-acute care for breakthrough infections.

Summary: This study demonstrates that SARS-CoV-2 can infect the eyes and cause retinal inflammation in transgenic mice and Syrian hamster models. The virus can also spread from the lungs to the brain and eyes through neuronal pathways. These findings contribute to a better understanding of viral transmission and clinical characteristics of SARS-CoV-2 and can aid in improving COVID-19 control procedures.

Platelet-derived extracellular vesicles to target plaque inflammation for effective anti-atherosclerotic therapy

Summary: Engineering platelet-derived extracellular vesicles (PEVs) to load MCC950, an NLRP3-inflammasome inhibitor, for atherosclerosis-targeted therapy can significantly reduce plaque formation, lower local inflammation, and inhibit proliferation of macrophages and T cells at the plaque site compared with free drug administration. This strategy indicates the promising potential of PEVs for targeted drug delivery in atherosclerosis treatment.

Attributes and predictors of long COVID

Carole H. Sudre et al.

Summary: Reports on long-lasting COVID-19 symptoms, known as 'long COVID', are increasing. Analysis of data from over 4,000 COVID-19 cases revealed that long COVID is more common in older individuals, those with higher body mass index, and females. Experiencing more than five symptoms in the first week of illness was associated with long COVID. A simple model showed promise in distinguishing between short and long COVID cases early in the disease.

Gut vascular barrier impairment leads to intestinal bacteria dissemination and colorectal cancer metastasis to liver

Summary: Bacteria from primary tumors can lead to bacterial dissemination from the intestine to the liver, disrupting the gut vascular barrier, boosting the formation of a premetastatic niche, and enhancing the recruitment of metastatic cells. High levels of PV-1, a marker of impaired gut vascular barrier, are associated with liver bacteria dissemination and metachronous distant metastases in colorectal cancer patients, making it a prognostic marker for distant recurrence and vascular impairment.

Neuroinflammatory astrocyte subtypes in the mouse brain

Philip Hasel et al.

Summary: This study investigated the transcriptome changes of mouse cortical astrocytes after an acute inflammatory stimulus, revealing rapid and drastic changes over time. By sequencing thousands of astrocytes at single-cell resolution, it was shown that inflammation causes a widespread response with distinct subtypes undergoing inflammatory transitions with defined transcriptomic profiles. Spatial transcriptomics and in situ hybridization were used to attribute specific inflammation-induced reactive astrocyte sub-states to certain brain regions.

Is There a Brain Microbiome?

Christopher D. Link

Summary: Numerous studies have identified microbial sequences or epitopes in pathological and non-pathological human brain samples, but it is still uncertain whether these observations are artifactual or truly representative of brain contamination by microbes. While the evidence for the presence of microbes in diseased brains is strong, a convincing demonstration of resident microbes in the healthy human brain is still lacking. Dedicated animal model studies may be necessary to determine the existence of a brain microbiome.
