APOBEC mutagenesis is a common process in normal human small intestine

Yichen Wang et al.

Summary: APOBEC mutational signatures SBS2 and SBS13 are commonly present in various human cancer types, but the stimulus, timing, and responsible APOBEC enzymes are not fully understood. Whole-genome sequencing of 342 microdissected normal epithelial crypts from the small intestines of 39 individuals revealed that SBS2/SBS13 mutations occurred in 17% of crypts, which is more frequent than in most other normal tissues. The study suggests that the high levels of SBS2/SBS13 in the small intestine are a result of APOBEC1 fulfilling its physiological function of editing APOB mRNA. Overall, APOBEC enzymes are a common contributor to the mutational burden in small intestine tissue.

Therapy-induced APOBEC3A drives evolution of persistent cancer cells

Hideko Isozaki et al.

Summary: Acquired drug resistance to anticancer targeted therapies remains a clinical challenge. This study reveals that lung cancer targeted therapies can induce expression of APOBEC3A (A3A), leading to sustained mutagenesis. Furthermore, it shows that A3A induction promotes the formation of double-strand DNA breaks, increasing genomic instability in drug-tolerant cancer cells.

Loss of SYNCRIP unleashes APOBEC-driven mutagenesis, tumor heterogeneity, and AR-targeted therapy resistance in prostate cancer

Xiaoling Li et al.

Summary: This study reveals that SYNCRIP acts as an endogenous molecular brake to suppress APOBEC-driven mutagenesis in prostate cancer, which contributes to tumor heterogeneity and therapy resistance. Overactivated APOBEC3B, in SYNCRIP-deficient prostate cancer cells, is identified as a key mutator that generates driver mutations in frequently mutated genes in prostate cancer, including FOXA1 and EP300. Functional screening identifies eight crucial driver genes (BRD7, CBX8, EP300, FOXA1, HDAC5, HSF4, STAT3, and AR) mutated by APOBEC3B, which confer resistance to androgen receptor-targeted therapy in prostate cancer.

Monoallelic deleterious MUTYH germline variants as a driver for tumorigenesis

Rodrigo Araujo Sequeira Barreiro et al.

Summary: Monoallelic pathogenic MUTYH germline variants may lead to tumorigenesis through somatic loss of heterozygosity of the functional MUTYH allele in the tumor. The frequency of monoallelic pathogenic MUTYH germline variants is higher in individuals with cancer than in the general population, although this frequency is not uniform among tumor types.

Mechanisms of APOBEC3 mutagenesis in human cancer cells

Mia Petljak et al.

Summary: The study reveals that endogenous APOBEC3 deaminases generate prevalent mutational signatures in human cancer cells, with APOBEC3A identified as the main driver of these mutations. APOBEC3B can restrain APOBEC3A-dependent mutagenesis to some extent while contributing its own smaller mutation burdens, and the mechanisms translating APOBEC3 activities into distinct mutational signatures are dissected.

Chromatin accessibility of primary human cancers ties regional mutational processes and signatures with tissues of origin

Oliver Ocsenas et al.

Summary: Somatic mutations in cancer genomes are associated with DNA replication timing and chromatin accessibility. This study uses machine learning to model mutation burden in cancer genomes and finds that the epigenomes of primary cancers are the strongest predictors of mutation burden. The genomic distributions of mutational signatures are also influenced by the epigenomes of matched cancer and tissue types. This study highlights the association of regional mutational processes with the epigenomes of primary cancers.

Structure-Guided Design of a Potent and Specific Inhibitor against the Genomic Mutator APOBEC3A

Juan C. Serrano et al.

Summary: Nucleic acid structure is crucial for the selectivity of DNA and RNA-modifying enzymes. In this study, a nucleic acid-based inhibitor of A3A was developed by embedding a mechanism-based inhibitor into a DNA dumbbell structure that mimics the ideal substrate secondary structure. The inhibitor shows potent inhibition and specificity against A3A.

Fat1 deletion promotes hybrid EMT state, tumour stemness and metastasis

Ievgenia Pastushenko et al.

Summary: This study reveals that mutation of the FAT1 gene promotes tumor initiation, progression, invasiveness, stemness, and metastasis. Loss of function of FAT1 activates a CAMK2-CD44-SRC axis while inactivates EZH2, playing important roles in the expression of YAP1 and ZEB1.

Loss of MGA repression mediated by an atypical polycomb complex promotes tumor progression and invasiveness

Haritha Mathsyaraja et al.

Summary: The transcription factor MGA, a member of the MYC network, plays a tumor suppressive role in vivo, as shown in mouse models of non-small cell lung cancer. Its loss accelerates tumor growth, enhances invasive capabilities, and affects the expression of genes associated with metastasis. MGA-MAX, E2F6, and L3MBTL2 co-occupy thousands of promoters, with MGA stabilizing these subunits for function.
