Amide/Ester Cross-Coupling via C-N/C-H Bond Cleavage: Synthesis of β-Ketoesters

Summary: The coupling of activated primary, secondary, and tertiary amides with enolizable esters in the presence of LiHMDS at room temperature yielded good yields of beta-ketoesters. This protocol provides an efficient, mild, and highly chemoselective method for the synthesis of beta-alkylketoesters through cross-coupling between aliphatic amides and esters. Additionally, gram-scale secondary and primary amides exhibited good reactivity when coupled with esters via in situ generated activated tertiary amides.

Iodine-Mediated Synthesis of 2-(Methylthio)-4H-chromen-4-ones and Study of Their Halogenation Reactions

Summary: An efficient iodine-mediated method has been developed for the synthesis of functionalized 2-(methylthio)-4H-chromen-4-ones, which serve as key precursors for various chemical reactions. Mechanistically, the intramolecular cyclization of ketene dithioacetal was observed to proceed through a radical pathway. 3-Halo-2-(methylthio)-4H-chromen-4-ones were obtained via various two- or one-pot halogenation approaches.
