Anti-proliferative potential of copper(I) acylthiourea complexes with triphenylphosphine against breast cancer cells

Summary: We synthesized five new Cu(I) acylthiourea complexes (C1-C5) and characterized them using analytical and spectroscopic tools. The complexes exhibited a distorted tetrahedral geometry as observed from a single crystal X-ray diffraction study. Complex C4 showed the highest binding efficacy with biomolecules and demonstrated high cytotoxicity against breast cancer cell lines. It induced apoptosis through a reactive oxygen species (ROS)-mediated mitochondrial signaling pathway.

Copper(I)-Thiosemicarbazone Complexes with Dual Anticancer and Antiparasitic Activity

Summary: Four new Cu(I) complexes were synthesized and characterized, and their anti-trypanosome and anticancer activities were evaluated. The complexes showed higher cytotoxicity against T. cruzi and chemoresistant prostate PC3 cells compared to benchmark drugs nifurtimox and cisplatin. They also exhibited high cellular internalization and activation of apoptosis in ovarian OVCAR3 cells. However, the production of reactive oxygen species was not significant.

Summary: Two heterocyclic azole compounds, SVS1 and SVS2, were unexpectedly synthesized from different starting materials. The structural and binding studies showed that these compounds exhibit comparable binding efficiency with SARS-CoV-2 and ACE-2 as established drugs, with SVS1 showing better docking energy.

Thiosemicarbazone Derivatives Developed to Overcome COTI-2 Resistance

Summary: In addition to zinc, iron and copper also play a role in the mode of action and resistance development of COTI-2 and its derivatives. COTI-NMe2 is discovered to be a new drug candidate with improved anticancer activity and resistance profile. The study reveals that COTI-2 forms stable complexes with copper and glutathione, making it a substrate for the resistance efflux transporter ABCC1. Furthermore, the interaction of COTI-2 and its derivatives with iron, copper, and zinc ions is characterized, and their activities against drug-resistant cancer cells are investigated.

Compared with its analogs 1D-Zn and OD-Zn, 2D-Zn shows the maximum alteration in the conformation of 2-cpa and exhibits the largest red-shift in the fluorescence emission peak.

Summary: In this study, two novel Ag(I)-based supramolecular structures were synthesized using bis(pyridyl) ancillary ligands to assist the main ligand pyrazine sulfonic acid, which improved the rigidity and coordination ability of the main ligand. The application of bis(pyridyl) ancillary ligands combined with weak interactions extended the structure to higher dimensions.
