A two-gene marker for the two-tiered innate immune response in COVID-19 patients

Hongxing M. Lei

Summary: A study on blood samples from COVID-19 patients found that IFI27 activation (a gene associated with viral infection) was not correlated with disease severity, while S100A12 activation (a gene associated with bacterial infection) was more common in severe patients. IFI27 activation was most pronounced within the first week of symptom onset and decreased after 2-3 weeks, while S100A12 activation showed no apparent correlation with disease duration and could last for several months. Understanding these two-tiered immune response features can contribute to our understanding of the disease mechanism and clinical triage of COVID-19. Developing a convenient two-gene protocol for monitoring these immune responses would be a valuable addition to current laboratory tests.

Advances in nucleic acid amplification techniques (NAATs): COVID-19 point-of-care diagnostics as an example

Tongjia Kang et al.

Summary: Achieving superhigh sensitivity is the ultimate goal in modern bio-detection. Nucleic acid amplification technologies (NAATs) are revolutionizing the field, offering novel methods for diagnosis with high efficiency and cost-effectiveness. This review discusses the principles, recent advancements, strengths, and challenges of different NAATs, with a focus on rapid and accurate point-of-care diagnosis. The application of these technologies in the context of the COVID-19 pandemic is also explored, emphasizing the significance of NAATs in combating the disease. Furthermore, the potential for enhancing sensitivity and specificity in biodetection through NAATs is discussed, aiming to fully exploit the capabilities of each technique.

A GMR-based assay for quantification of the human response to influenza

Neeraja Ravi et al.

Summary: Detecting and quantifying the host transcriptional response to influenza virus infection can serve as a real-time diagnostic tool for clinical management. We have developed a novel assay based on the influenza metasignature (IMS) using GMR sensors, which can classify influenza infection based on transcript levels. We also validated the accuracy of a single biomarker in stratifying patients with influenza.

From saliva to SNP: non-invasive, point-of-care genotyping for precision medicine applications using recombinase polymerase amplification and giant magnetoresistive nanosensors

Ana Sofia de Olazarra et al.

Summary: Genetic testing is vital for precision medicine, but the lack of affordable and widely deployable genotyping tools hinders its integration into routine clinical practice. This study presents a portable and automated platform using RPA and GMR technology for SNP genotyping at point-of-care. The results showed 100% accordance with traditional genotyping methods, demonstrating the potential of this approach for SNP genotyping in clinical settings.

IFI27 may predict and evaluate the severity of respiratory syncytial virus infection in preterm infants

Junyan Gao et al.

Summary: This study identified differentially expressed genes associated with RSV infection susceptibility in preterm infants and found IFI27 as a potential biomarker for predicting and evaluating the severity of RSV infection. High expression of IFI27 was correlated with more severe RSV cases, increased need for mechanical ventilation, more frequent hospitalizations, and longer cumulative hospital stays.

Ultrasensitive and Highly Specific Lateral Flow Assays for Point-of-Care Diagnosis

Yilin Liu et al.

Summary: LFAs, paper-based point-of-care diagnostic tools, have traditionally lower sensitivities and specificities than standard laboratory tests. Recent efforts have focused on optimizing assay kinetics, signal amplification, and sample preamplification to increase sensitivities, as well as improving specificity through high-affinity molecules and assay optimization. With continuing improvements, LFAs may soon offer performance competitive with laboratory techniques while retaining a rapid format.
