Preservation of the naive features of mesenchymal stromal cells in vitro: Comparison of cell- and bone-derived decellularized extracellular matrix

Ana Rita Pereira et al.

Summary: The fate and behavior of BM-MSC are influenced by their microenvironment. In this study, four different 2D cell-based matrices and a 3D decellularized human trabecular-bone scaffold were used to evaluate BM-MSC behavior. The cell-derived matrices were reliable for analyzing osteogenic differentiation, while the decellularized bone scaffold showed selective expression of bone-specific proteins. The complex architecture and physico-chemical properties of the scaffold upregulated stemness associated genes, indicating the conservation of naive features of BM-MSC in vitro.

Synergy effects of Asperosaponin VI and bioactive factor BMP-2 on osteogenesis and anti-osteoclastogenesis

Fangping Chen et al.

Summary: This study systematically investigated the roles of Chinese medicine Asperosaponin VI (ASP VI) on osteogenic mineralization and osteoclastogenesis. The results showed that ASP VI promotes osteogenesis and inhibits osteoclastic differentiation. Furthermore, the combination of ASP VI and BS enhances osteogenesis, promotes angiogenesis, and suppresses osteoclastogenesis. These findings provide new insights into the care and prevention of osteoporosis.

Phosphorylation inhibition of protein-tyrosine phosphatase 1B tyrosine-152 induces bone regeneration coupled with angiogenesis for bone tissue engineering

Yong Tang et al.

Summary: The phosphorylation state of PTP1B Y152 plays a central role in initiating the departure of cells from their niches during bone regeneration. A peptide called 152RM was found to promote cell migration via integrin avp3, enhance MSCs osteogenic differentiation by inhibiting ATF3, and induce type H vessels formation by activating Notch signaling. In vivo studies showed that DBM-MSN/152RM scaffolds were effective in inducing bone formation and type H vessels expansion.
