MYB30 and MYB14 form a repressor-activator module with WRKY8 that controls stilbene biosynthesis in grapevine

Huayuan Mu et al.

Summary: When exposed to pathogen infection or UV radiation, grapevine plants rapidly accumulate the stilbenoid resveratrol with concomitant increase of stilbene synthase (STS). The VvMYB14-VvWRKY8-VvMYB30 regulatory circuit plays a key role in responding to UV stress and fine-tuning stilbene biosynthesis in grapevine. This study reveals the complex regulatory network involved in stilbene biosynthesis in grapevine.

Dissecting the labdane-related diterpenoid biosynthetic gene clusters in rice reveals directional cross-cluster phytotoxicity

Riqing Li et al.

Summary: This study used CRISPR/Cas9 technology to precisely delete two biosynthetic gene clusters related to labdane-related diterpenoids in rice, showing that deletion of one cluster leads to a lesion mimic phenotype while deletion of the other does not. This demonstrates directional cross-cluster interactions between these two gene clusters.

Characterization of a Group of UDP-Glycosyltransferases Involved in the Biosynthesis of Triterpenoid Saponins of Panax notoginseng

Yanting Li et al.

Summary: This study established an efficient heterologous expression system for evaluating the UGT-mediated glycosylation process of triterpene saponins. Six UGTs were identified and shown to be responsible for the glycosylation reactions of 21 triterpenoid saponins. UGTPn87 exhibited promiscuous sugar-donor specificity and generated two new saponins. The biosynthetic pathways of 17 saponins were elucidated, revealing the presence of four new saponins in Panax for the first time.

Overexpression of PnMYB2 from Panax notoginseng induces cellulose and lignin biosynthesis during cell wall formation

Yue Shi et al.

Summary: The transcription factor PnMYB2 in Panax notoginseng plays a crucial role in the formation of cell wall by promoting the synthesis of cellulose and lignin. Its overexpression in Arabidopsis thaliana leads to increased thickness of cell wall due to enhanced synthesis of cellulose and lignin. This study provides insights into the mechanism of cell wall formation in plants.

Structural and interactions analysis of a transcription factor PnMYB2 in Panax notoginseng

Pengguo Xia et al.

Summary: This study identified and characterized the interaction between PnMYB2 and key enzymes in Panax notoginseng. The study also confirmed the interaction between PnMYB2 and the upstream sequences of PnSS and PnSE1 promoters. The full-length cDNA sequence of PnMYB2 was isolated and its structure and phylogenetic analysis were conducted. The study showed that PnMYB2 is localized in the nucleus and can specifically bind to certain elements. This research provides a theoretical basis for the transcriptional regulation of saponin biosynthesis in Panax notoginseng.

Interplay between R2R3 MYB-type activators and repressors regulates proanthocyanidin biosynthesis in banana (Musa acuminata)

Ruchika Rajput et al.

Summary: This study characterized the transcriptional regulatory network of proanthocyanidin biosynthesis in banana. The researchers identified two activators and four repressors that fine tune the biosynthesis of proanthocyanidins. The activators interact with other proteins to transactivate the genes involved in proanthocyanidin biosynthesis, while the repressors form complexes with these proteins to repress proanthocyanidin accumulation. Moreover, the study revealed a competitive relationship between the activators and the repressors, providing insights into the agile regulation of proanthocyanidin biosynthesis.

The antagonistic MYB paralogs RH1 and RH2 govern anthocyanin leaf markings in Medicago truncatula

Chongnan Wang et al.

Summary: The study identified two R2R3 MYB paralogous regulators, RH1 and RH2, in determining anthocyanin leaf markings in Medicago truncatula. RH1 acts as an activator for anthocyanin biosynthesis, while RH2 competes with RH1 in the regulatory complex and has opposite effects. RH1 can activate its own expression, while RH2-mediated competition can suppress RH1 expression, revealing a multidimensional paralogous-antagonistic regulatory paradigm for fine-tuning patterned pigmentation.

An HD-ZIP-MYB complex regulates glandular secretory trichome initiation in Artemisia annua

Lihui Xie et al.

Summary: The study identified key transcription factors, AaMYB16 and AaMYB5, that interact with AaHD1 to regulate glandular secretory trichomes (GSTs) initiation in plants. AaMYB16 was found to be a positive regulator while AaMYB5 had an inhibitory effect on GST initiation. The competitive interaction between AaMYB16 and AaMYB5 modulates the binding activity of AaHD1 promoter, providing new insights into the regulatory network controlling GST formation.
