Novel variants in ZP1, ZP2 and ZP3 associated with empty follicle syndrome and abnormal zona pellucida

Liwei Sun et al.

Summary: Whole-exome sequencing was performed in probands with empty follicle syndrome (EFS) and abnormal zona pellucida (ZP), revealing a homozygous nonsense variant in ZP1 associated with EFS, a homozygous frameshift variant in ZP2 triggering protein degradation, and a heterozygous missense variant in ZP3 linked to ZP morphological abnormalities. These findings expand the mutational spectrum of ZP1, ZP2, and ZP3, and provide new evidence for genetic diagnosis of female infertility.

ZP3 and AIPL1 participate in GVBD of mouse oocytes by affecting the nuclear membrane localization and maturation of farnesylated prelamin A

Zhen Jin et al.

Summary: The low maturation rate of oocytes is a significant factor in female infertility and assisted pregnancy failure. This study investigates the mechanism of zona pellucida 3 (ZP3) and lamin A in oocyte maturation, suggesting that stabilizing prelamin A promotes germinal vesicle breakdown (GVBD).

Summary: A novel mutation in ZP3 associated with empty follicle syndrome was identified, leading to infertility in mice and impacting ZP formation. The study suggests that a loop structure in ZP3 plays a critical role in specifying the correct heterodimerization partner.

Summary: This study investigates the association between a recurrent heterozygous mutation in ZP2 and Empty follicle syndrome (EFS). The results suggest that this mutation is associated with female infertility and leads to ZP2 accumulation in the cytoplasm of oocytes.

Novel mutations in ZP2 and ZP3 cause female infertility in three patients

Weimin Jia et al.

Summary: The aim of this study was to identify disease-causing mutations in three infertile female patients diagnosed with abnormal zona pellucida and empty follicle syndrome. The study found three novel mutations in the ZP2 and ZP3 genes, expanding the spectrum of ZP gene mutations and phenotypes, which is beneficial for the genetic diagnosis of infertility in females.

Ultrasensitive Ribo-seq reveals translational landscapes during mammalian oocyte-to-embryo transition and pre-implantation development

Zhuqing Xiong et al.

Summary: Researchers used low-input Ribo-seq technology in combination with PAIso-seq to investigate the translational landscape during oocyte-to-embryo transition in mammals. They found that translation activation is closely associated with polyadenylation, while translation repression is similar to global de-adenylation.

New Insights into the Mammalian Egg Zona Pellucida

Carla Moros-Nicolas et al.

Summary: The zona pellucida (ZP) is a ancient extracellular matrix surrounding mammalian oocytes and embryos, preventing polyspermy and protecting the embryo. Studies have shown differences in ZP composition among different mammalian species, likely due to molecular evolution of the ZP gene family. Interactions with other proteins in the oviductal lumen may play different roles in different mammalian species.

Zona Pellucida Genes and Proteins: Essential Players in Mammalian Oogenesis and Fertility

Paul M. Wassarman et al.

Summary: The zona pellucida (ZP) surrounding mammalian oocytes and eggs consists of unique glycosylated proteins (ZP1-4), with polypeptide regions (ZPD) that play a crucial role in the polymerization of ZP proteins into fibrils. ZP genes are conserved among mammals and regulated by cis-acting sequences and trans-acting factors. Mutations in ZP genes can detrimentally affect ZP formation and female fertility, highlighting the importance of ZPD subdomains in fibril polymerization.
