Regiodivergent Metal-Controlled Synthesis of Multiply Substituted Quinolin-2-yl- and Quinolin-3-ylphosphonates

Summary: In this study, a selective method for synthesizing multi-substituted quinoline-2-ylphosphonates and quinoline-3-ylphosphonates was developed using copper or gold-catalyzed reactions. The reactions involved phosphoryl-substituted conjugated ynones and 2'-amino-2,2,2-trifluoroacetophenones, resulting in various substituted quinolines with good yields. Additionally, it was discovered that (4,4,4-trifluoro-3-oxobut-1-yn-1-yl)phosphonate reacted with 2-aminoaryl ketones under non-catalytic conditions, leading to the formation of 4-substituted quinoline-2-ylphosphonates in high yields.

Post-Ugi Cyclizations Towards Polycyclic N-Heterocycles

Summary: The Ugi reaction is highly explored due to its mild reaction conditions, wide scope, and high variability for the formation of multifunctional adducts. By carefully selecting the starting four components, Ugi-adducts can undergo different post-transformations to synthesize bioactive heterocycles, natural products, and macrocycles. Various post-Ugi transformations have been developed for constructing structurally novel polycycles, and this account summarizes important efforts for the synthesis of polycyclic N-heterocycles via post-Ugi cyclizations from the Van der Eycken laboratory onwards 2016. With the aid of transition metal catalysis and metal-free strategies, versatile polyheterocycles are prepared with high efficiency and step-economy.

Recent Access to Polycycles via Post-Ugi Reactions

By carefully choosing the starting four components, Ugi adducts have proven to be highly efficient for various post-transformations. Various post-Ugi transformations have been employed for the synthesis of structurally novel polycyclic N-heterocycles. This minireview summarizes the recent developments of post-Ugi reactions for the synthesis of polycycles after the year 2019, utilizing transition metal catalysis and transition metal-free approaches.

This review provides an overview of recent advances in the preparation of isoindolo[2,1-b]isoquinolin-5(7H)-ones using transition metal-catalyzed C-H activation/annulation, aiming to help researchers discover hidden opportunities and accelerate the discovery of novel transformations based on C-H activation/annulation.

Gold(I)-Catalyzed Intramolecular Bicyclization: Divergent Construction of Quinazolinone and Ampakine Analogues

Summary: A gold(I)-catalyzed cascade intramolecular cyclization of Ugi adducts has been developed for the divergent construction of quinazolinone and ampakine analogues in a rapid, efficient and step-economical manner. This method shows high yields, excellent functional-group tolerance, broad substrate scope and excellent chemo- and regioselectivity. The practicality of this strategy is further demonstrated by a scale-up reaction. Diverse oligopeptides are also found to be well tolerated, affording polycyclic products decorated with a peptide chain.

Cationic Co(I) Catalysts for Regiodivergent Hydroalkenylation of 1,6-Enynes: An Uncommon cis-β-C-H Activation Leads to Z-Selective Coupling of Acrylates

Summary: Two intermolecular hydroalkenylation reactions of 1,6-enynes were demonstrated, leading to the formation of substituted 5-membered carbo- and heterocycles. The interactions of alkenes with a cobaltacyclopentene intermediate were found to be crucial, with unexpected beta-C-H activation observed in the case of acrylates. The reaction rate was dependent on the concentrations of cobalt, acrylate, and activator, indicating a complex mechanism involving multiple factors.
