The developed protocol is equally efficient for homocoupling enamines to form symmetric pyrroles.

Biocatalytic One-Carbon Ring Expansion of Aziridines to Azetidines via a Highly Enantioselective [1,2]-Stevens Rearrangement

Summary: The study reports the enantioselective one-carbon ring expansion of aziridines to form azetidines through engineered carbene transferase enzymes. A variant of cytochrome P450BM3, P411-AzetS, showed unparalleled stereocontrol (99:1 er) in the [1,2]-Stevens rearrangement and could override the inherent reactivity of aziridinium ylides to perform this rearrangement. By controlling the reactive aziridinium ylide intermediates, these evolvable biocatalysts enable a transformation not achievable by other catalyst classes.

Summary: Atropisomers beyond the C-C axis have become important additions to the field of axially chiral compounds. Previously considered challenging, these atropisomers characterized by C-N, C-O, C-B, or N-N bonds have been found to possess stable axially chiral frameworks due to their shorter bond lengths and electron-repelling effects. Recent years have seen rapid progress in this emerging area, with various catalytic approaches being reported for their efficient synthesis. These X-Y axially chiral compounds are valuable molecules that can be utilized as ligands or catalysts in asymmetric catalysis or evaluated for their potential biological activities. The chemistry of atropisomers beyond C-C axial chirality is believed to hold an important position in organic chemistry and beyond.

Aldehyde-catalysed carboxylate exchange in α-amino acids with isotopically labelled CO2

However, the labelling of carbon-labelled alpha-amino acids is difficult and time-consuming, resulting in high cost and scarcity of C-labelled products. This study demonstrates that aldehydes can catalyse the isotopic carboxylate exchange of alpha-amino acids with *CO2, providing a new method for late-stage C-11 radiolabelling.

The Rational Design and Atroposelective Synthesis of Axially Chiral C2-Arylpyrrole-Derived Amino Alcohols

Summary: In this study, a versatile C-1-symmetric biaryl amino alcohol scaffold based on axially chiral C2-arylpyrrole framework was designed and synthesized. A chiral phosphoric acid-catalyzed reaction method was established to obtain NPNOL compounds with high yields and high atroposelectivity. DFT calculations revealed the reaction mechanism and the origin of enantioselectivity. The NPNOL-derived products could be easily transformed into organocatalysts/ligands and showed promising utility in asymmetric catalytic reactions.

Enantioselective Synthesis of N-N Bisindole Atropisomers

Summary: The first enantioselective synthesis of N-N bisindole atropisomers via palladium-catalyzed construction of one indole skeleton is reported in this study. A wide variety of N-N axially chiral bisindoles were generated with good yields and excellent enantioselectivities through a cascade condensation/N-arylation reaction. The reaction mechanism and enantiocontrol were further investigated using density functional theory (DFT) calculations.

Direct Deamination of Primary Amines via Isodiazene Intermediates

Summary: The study reports a reaction that selectively deaminates primary amines and anilines under mild conditions with remarkable functional group tolerance, including pharmaceutical compounds, amino acids, amino sugars, and natural products. An anomeric amide reagent facilitates the reaction through the intermediacy of an unprecedented monosubstituted isodiazene intermediate, simplifying deamination and enabling strategic applications of iminium and amine-directed chemistries as traceless methods. Mechanistic and computational studies support the intermedicacy of a primary isodiazene, which exhibits unexpected divergence from previously studied secondary isodiazenes, leading to cage-escaping, free radical species undergoing a chain hydrogen-atom transfer process with aliphatic and diazenyl radical intermediates.
