Solute exchange through gap junctions lessens the adverse effects of inactivating mutations in metabolite-handling genes

Summary: The growth of cancer cells can be slowed down by deactivating certain metabolic pathways. However, loss-of-function mutations in metabolic pathways are not negatively selected in human cancers, which suggests that these genes are not necessary for cancer cell growth in vivo. It is hypothesized that spontaneous mutations in metabolic genes may not lead to functional defects as mutated cells can be rescued by exchanging metabolites with neighboring wild-type cells through gap junctions.
