Epitranscriptomic regulation of cortical neurogenesis via Mettl8-dependent mitochondrial tRNA m3C modification

Feng Zhang et al.

Summary: Increasing evidence supports the crucial roles of METTL8 and mitochondrial tRNA modification in maintaining embryonic cortical neural stem cells in mice and humans. Deletion of METTL8 leads to reduced mitochondrial protein synthesis and attenuated respiration, resulting in impaired neural stem cell maintenance. These findings highlight the conserved epitranscriptomic mechanism of METTL8 in regulating mitochondrial function and neural stem cell homeostasis.

Roles and dynamics of 3-methylcytidine in cellular RNAs

Katherine E. Bohnsack et al.

Summary: Modified nucleotides within cellular RNAs play significant roles in their biogenesis, stability, and function. The recent discovery of the methyltransferases responsible for installing 3-methylcytidine (m(3)C(32)) in human tRNAs has shed light on the importance of this modification. Mechanistic details of how these methyltransferases recognize their substrate tRNAs have been uncovered, along with the biological and functional relevance of the interconnections between m(3)C(32) and modified adenosines at position 37. Functional insights suggest that m(3)C(32) modifications influence tRNA structure and impaired translation is observed in the absence of these modifications. The development of quantitative, transcriptome-wide m(3)C mapping approaches and the discovery of an m(3)C demethylase indicate the dynamic nature of m(3)C, potentially contributing to fine-tuning gene expression in different conditions.

Selective degradation of tRNASer(AGY) is the primary driver for mitochondrial seryl-tRNA synthetase-related disease

Tingting Yu et al.

Summary: Mitochondrial translation is crucial for cellular energy homeostasis, and variations in mitochondrial aaRSs can lead to various human diseases. This study identified two novel variants of SARS2 that cause a multisystem disorder. These variants exhibited reduced tRNA binding and aminoacylation capacities, resulting in mitochondrial dysfunction and decreased tRNA abundance due to RNA degradation. The findings highlight the importance of reduced tRNA(Ser)(AGY) abundance in the development of SARS2-related diseases.

Gui-Xin Peng et al.

Summary: The study found that mitochondrial aminoacyl-tRNA synthetases (mito aaRSs) partially colocalize with mitochondrial RNA granules (MRGs), possibly facilitated by their tRNA-binding capacity. Additionally, several mito aaRS-containing complexes were discovered, and their activities were influenced by interactions. These findings deepen our understanding of the functional and regulatory mechanisms of mito aaRSs.
