Role of alarmins in poststroke inflammation and neuronal repair

Summary: Severe loss of cerebral blood flow leads to hypoxia and glucose deprivation, causing cell death in the ischemic brain. Dead cells release alarmins to activate immune cells, resulting in inflammation and neuronal cell death. Poststroke inflammation resolves within several days, and neural repair restores neurological functions. Clearance of damaged brain molecules is crucial for resolving inflammation. This review describes the molecular dynamics, signaling pathways, and functions of alarmins in poststroke inflammation and resolution.

Estimated Burden of Stroke in China in 2020

Extracellular CIRP dysregulates macrophage bacterial phagocytosis in sepsis

Summary: In this study, we identified a novel mechanism by which eCIRP impairs macrophage bacterial phagocytosis in sepsis. Increased eCIRP levels were associated with decreased bacterial clearance in infected mice. eCIRP inhibited actin remodeling in macrophages through the formation of a STAT3-beta PIX complex, resulting in phagocytic dysfunction. Inhibition of STAT3 rescued the macrophage phagocytic dysfunction induced by eCIRP.

Summary: Sterile inflammation caused by DAMPs aggravates the prognosis of primary injury such as ischemic stroke. The AIM protein can reduce sterile inflammation by attenuating DAMPs and administration of AIM can ameliorate the deleterious effects of ischemic stroke. AIM binds to DAMPs through charge-based interactions and disulfide bond formation, promoting their phagocytic removal and neutralizing their inflammatory effects.AIM-deficient mice in experimental stroke show severe damage and higher mortality, while administration of recombinant AIM reduces sterile inflammation in the infarcted region, leading to a significant reduction in animal mortality. Targeting DAMPs may improve therapies for ischemic stroke.
