Sporothrix globosa melanin regulates autophagy via the TLR2 signaling pathway in THP-1 macrophages

Summary: Melanin, an important virulence factor of pathogenic fungi, has been shown to suppress host immune responses. However, its potential influence on autophagy, a vital cellular mechanism underlying the host's innate immunity, has not been explored. This study investigated the effect of melanin on autophagy in macrophages and revealed the novel immune defense ability of Sporothrix globosa melanin to inhibit macrophage functionality through the regulation of Toll-like receptor (TLR) expression.

Summary: Talaromycosis is an invasive mycosis caused by Talaromyces marneffei, and it is endemic in tropical and subtropical Asia. The infection rate is high, with approximately 17,300 cases diagnosed annually, and the mortality rate is extremely high. However, despite its impact on immunocompromised individuals and the increase in cases among those without HIV, global attention to diagnostic and therapeutic approaches for talaromycosis has been lacking. Understanding the pathogenic mechanism of T. marneffei has been limited, but recent research breakthroughs have shed light on its growth biology and interaction with the host immune response and environmental stimuli.
