Genome-Wide mRNA and Long Non-Coding RNA Analysis of Porcine Trophoblast Cells Infected with Porcine Reproductive and Respiratory Syndrome Virus Associated with Reproductive Failure

Xinming Zhang et al.

Summary: This study investigated the differential expression of mRNA and lncRNA in porcine trophoblast cells infected with PRRSV. It was found that PRRSV inhibits the GPER1-PI3K-AKT-mTOR signaling pathway, leading to disturbances in cell proliferation, differentiation, and nutrient metabolism. PRRSV also induced trophoblast cell apoptosis and disrupted the integrity of the placental villus barrier.

PRRSV-1 induced lung lesion is associated with an imbalance between costimulatory and coinhibitory immune checkpoints

Ines Ruedas-Torres et al.

Summary: This study evaluated the expression of immune checkpoints in the lungs and tracheobronchial lymph nodes of piglets infected with two different virulent strains of PRRSV-1 during the early stage of infection. The results showed that PRRSV infection dysregulates the immune response and leads to changes in the expression of costimulatory and coinhibitory molecules. The simultaneous expression of coinhibitory immune checkpoints may work together to control and limit inflammation-induced tissue damage.

Immune Molecules' mRNA Expression in Porcine Alveolar Macrophages Co-Infected with Porcine Reproductive and Respiratory Syndrome Virus and Porcine Circovirus Type 2

Zhiying Cui et al.

Summary: This study aimed to explore the pathogenesis mechanism of co-infection with PRRSV and PCV2. The results showed that co-infection had no effect on promoting PCV2 replication, but it could promote PRRSV replication. The immune regulatory molecules were down-regulated, while inflammatory factors and immune checkpoint molecules were up-regulated in the co-infection groups. These findings partially explain the enhanced pulmonary lesions caused by dual infection with PCV2 and PRRSV in PAMs.

Integrated time-series transcriptomic and metabolomic analyses reveal different inflammatory and adaptive immune responses contributing to host resistance to PRRSV

Qingqing Wu et al.

Summary: This study analyzed the transcriptomic and metabolomic responses of white blood cells and serum to PRRSV infection, revealing the differences in immune response and susceptibility mechanism. The study found that PRRSV infection regulates the gene expression levels of various immune cells, with more severe suppression of T cell immunity and inflammatory response in susceptible pigs. In addition, PRRSV infection also affects amino acid pathways and lipid metabolism. These findings provide a basis for understanding the mechanism of PRRSV resistance and effective control methods.

Unveiling the long non-coding RNA profile of porcine reproductive and respiratory syndrome virus-infected porcine alveolar macrophages

Junxin Gao et al.

Summary: This study identified a total of 350 annotated lncRNAs and 1792 novel lncRNAs in PAMs through RNA-seq analysis, with 86 differentially expressed lncRNAs and 406 DE protein-coding mRNAs identified upon PRRSV incubation. GO category and KEGG pathway enrichment analyses revealed that these DE lncRNAs and mRNAs were mainly involved in inflammation- and pathogen infection-induced pathways. The co-expression analysis demonstrated that several lncRNAs are positively correlated with their predicted target genes, suggesting that porcine lncRNAs play a role in regulating immune responses against PRRSV infection.

Up-Regulation of Immune Checkpoints in the Thymus of PRRSV-1-Infected Piglets in a Virulence-Dependent Fashion

Ines Ruedas-Torres et al.

Summary: This study aimed to evaluate the expression of selected immune checkpoints in the thymus of piglets infected with two different PRRSV-1 strains. The up-regulation of PD1/PDL1, CTLA4, TIM3, LAG3 and IDO1 immune checkpoints in the thymus of PRRSV-infected piglets, especially those infected with the virulent Lena strain from 6 dpi onwards, was associated with disease progression, high viral load and cell death.
