Comprehensive Analysis of an Individualized Immune-Related lncRNA Pair Signature in Gastric Cancer

Chuang Nie et al.

Summary: A personalized signature based on immune-related long noncoding RNAs (lncRNAs) was developed to predict the survival outcomes and immunotherapeutic responses of gastric cancer (GC) patients. The signature showed good applicability in discriminating the prognosis of pan-cancer patients. High-risk patients had lower survival rates due to aggressive clinical features, enriched cancer-related signaling pathways, specific immunosuppressive cell infiltration, and low tumor mutation burden. A nomogram was constructed and validated for more accurate predictions of GC prognosis.

Loss of cell division cycle-associated 5 promotes cell apoptosis by activating DNA damage response in clear cell renal cell carcinoma

Xing Huang et al.

Summary: CDCA5, highly expressed in ccRCC, is associated with poor clinical outcomes and distant metastasis risk. Knockdown of CDCA5 inhibits ccRCC growth and migration by inducing DNA damage and cell cycle arrest. This suggests that CDCA5 inhibition may improve outcomes in high-risk ccRCC patients.

Summary: This study aimed to evaluate the impact of DNA repair-related gene signature (DRGS) on the prognosis of colorectal cancer (CRC) patients. Through the analysis of gene expression profiles, a DRGS consisting of 11 genes was constructed to stratify patients into high- and low-risk groups. The results showed that patients in the high-risk group had worse disease-free survival. The validation of the model suggested that the low-risk group had better response to chemotherapy drugs and immune therapy.

Summary: This study assessed the prognostic potential of DNA damage repair markers in skin cutaneous melanoma by analyzing gene expression profiles from multiple databases. A prognostic signature consisting of twelve DNA repair-related genes was established, showing strong predictive ability for disease-specific survival. The signature was found to be associated with immune response, infiltrating immune cells, and sensitivity to chemotherapy and immune checkpoint inhibitors. Results provide insights for forecasting skin cutaneous melanoma progression and enhancing clinical benefits from immunotherapy.

A DCS-related lncRNA signature predicts the prognosis and chemotherapeutic response of patients with gastric cancer

Yang Zhang et al.

Summary: The aim of this study was to identify DCS mRNAs-lncRNAs associated with chemotherapy response and prognosis in gastric cancer patients. A prognostic signature including 11 lncRNAs was constructed using LASSO regression. The signature was validated and found to be an effective prognostic factor, and it was also able to predict chemotherapy drug response and immune infiltration in GC patients. Furthermore, knockdown of LINC00106 or UBE2R2-AS1 enhanced the proliferation and migration of GC cells in vitro. Overall, this DCS therapy-related lncRNA signature may serve as a new strategy for predicting chemotherapy response and prognosis in GC patients, and LINC00106 and UBE2R2-AS1 may have a tumor suppressive function in GC.

LncRNA linc00312 suppresses radiotherapy resistance by targeting DNA-PKcs and impairing DNA damage repair in nasopharyngeal carcinoma

Zhen Guo et al.

Summary: Our study found that linc00312 is significantly down-regulated in NPC tissues and higher expression of linc00312 is associated with longer overall survival and better short-term radiotherapy efficacy. Overexpression of linc00312 increases NPC cells' sensitivity to ionizing radiation by hindering the DNA repair and cell cycle control mechanisms. DNA-PKcs was identified as the binding protein of linc00312, revealing a novel mechanism in the DNA damage response.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
