Complement Inhibition in Myasthenia Gravis and Neuromyelitis Optica Spectrum Disorder

Dubravka Dodig et al.

Summary: This paper briefly reviews the role of complement activation in the pathogenesis of myasthenia gravis (MG) and neuromyelitis optica spectrum disorders (NMOSD) and discusses the rationale and evidence for complement inhibition as a method to manage these diseases. It also provides a Canadian perspective on the use of complement inhibition therapy through real-world cases of MG and NMOSD.

Summary: This study investigated clinical information of myasthenia gravis patients aged 50 and older, finding that comorbidities were common in both early-onset and late-onset patients, increasing with age but not differing significantly between the two groups.

Epidemiology, diagnostics, and biomarkers of autoimmune neuromuscular junction disorders

Anna Rostedt Punga et al.

Summary: Autoimmune neuromuscular junction disorders are rare, with myasthenia gravis being increasingly recognized in older individuals. Various diagnostic methods, such as serological antibody, electrophysiological, imaging, and pharmacological tests, are available for these disorders. Determining if the disease is paraneoplastic is important for prognosis. However, objective biomarkers are needed to predict the disease course of myasthenia gravis and Lambert-Eaton myasthenic syndrome.

Thymus Functionality Needs More Than a Few TECs

Pratibha Bhalla et al.

Summary: The thymus, a primary lymphoid organ, plays a crucial role in producing T cells of the immune system. It can be damaged under certain types of systemic stresses and undergoes functional decline during aging. The microenvironment of the thymus, established through interactions between various cell types, influences thymus functions. Understanding the contributions of different stromal cell types in the thymus can inform strategies for restoring thymus functionality.

Identification of genetic risk loci and prioritization of genes and pathways for myasthenia gravis: a genome-wide association study

Summary: This study identified genetic risk loci associated with myasthenia gravis through a genome-wide association study and demonstrated different genetic risk factors between early-onset and late-onset cases. Genetic correlation analysis also revealed a genetic link between myasthenia gravis and other autoimmune diseases.

Role of complement and potential of complement inhibitors in myasthenia gravis and neuromyelitis optica spectrum disorders: a brief review

Jayne L. Chamberlain et al.

Summary: The complement system is a key player in the innate immune system, providing protection against pathogens but potentially leading to autoimmune diseases when overactive. A cascade of complement proteins and regulators exist to regulate a proportionate response to pathogens. Though involved in driving the pathogenesis of various diseases, including NMOSD and MG, the complement system also presents abundant molecular targets for therapeutic intervention.

Summary: This case report describes a patient with thymoma-associated autoimmune encephalitis and positive Titin antibodies, who showed improvement after immunotherapy but later experienced recurrence post-thymectomy. The findings suggest that Titin antibodies may play a role in the treatment and prognosis of autoimmune encephalitis.
