KMT2D acetylation by CREBBP reveals a cooperative functional interaction at enhancers in normal and malignant germinal center B cells

Summary: Heterozygous inactivating mutations of KMT2D and CREBBP are common in B cell lymphoma, and they co-occur in a significant number of follicular lymphoma and EZB/C3 diffuse large B cell lymphoma cases. The combination of haploinsufficiency of Crebbp and Kmt2d promotes the expansion of abnormally polarized germinal centers, which is an early event in lymphoma development. These findings highlight the functional and biochemical interaction between Crebbp and Kmt2d, and its implications for the treatment of lymphomas with enhancer defects induced by their combined loss.

Tracing Founder Mutations in Circulating and Tissue-Resident Follicular Lymphoma Precursors

Summary: Follicular lymphomas (FL) are characterized by BCL2 translocations, often detectable in blood years before FL diagnosis, but also observed in aging healthy individuals, suggesting additional lesions are required for lymphomagenesis. CREBBP KAT domain mutations were the most commonly observed precursor lesions, distinguishing patients developing FL from healthy adults with or without BCL2 rearrangements. These mutations can be detected years before clinical diagnosis and may help discriminate individuals at risk for lymphoma development.

HX009, a novel BsAb dual targeting PD1 x CD47, demonstrates potent anti-lymphoma activity in preclinical models

Summary: Both PD1/PD-L1 and CD47 blockades have limited activity in most subtypes of NHL, except for NK/T-cell lymphoma. The hemotoxicity of anti-CD47 agents in clinic may explain their limitations. In this study, a bispecific antibody HX009 was developed, targeting PD1 and CD47, with weakened CD47 binding to reduce toxicity. In vitro and in vivo experiments demonstrated the antitumor activity of HX009, especially in lymphomas with upregulated CD47. These findings support further clinical development of HX009 for NHL treatment.

Tumor-activated lymph node fibroblasts suppress T cell function in diffuse large B cell lymphoma
