Glyphosate and Fluoride in High-Hardness Drinking Water Are Positively Associated with Chronic Kidney Disease of Unknown Etiology (CKDu) in Sri Lanka

Dietary heavy metal(loid)s exposure and prevalence of chronic kidney disease of unknown aetiology (CKDu) in Sri Lanka

Summary: Research in Sri Lanka has shown that intake of heavy metals and metalloids through diet, particularly from rice, vegetables, and fish, exceeds recommended daily limits in areas with high prevalence of CKDu. The entire population in the study area faces a risk of high exposure to lead, with no significant difference in estimated daily intake between different age and gender categories. Immediate interventions are needed to reduce dietary exposure to heavy metals and metalloids.

Significance of Mg-hardness and fluoride in drinking water on chronic kidney disease of unknown etiology in Monaragala, Sri Lanka

Summary: The study conducted in the Monaragala district of Sri Lanka revealed a CKDu prevalence of about 6.7%. Analysis of groundwater samples showed significant differences in the concentrations of certain ions between CKDu-prevalent locations and control locations. Higher levels of alkali and alkaline earth cations, as well as fluoride, were found in the drinking water sources used by CKDu patients, potentially relating to the disease.

Urinary biomarkers indicate pediatric renal injury among rural farming communities in Sri Lanka

Summary: This study investigated the early detection of renal abnormalities in selected pediatric communities in Sri Lanka using kidney injury molecule (KIM-1) and neutrophil gelatinase-associated lipocalin (NGAL). The results showed significantly elevated levels of urinary KIM-1 expression in children from CKDu affected regions, indicating low-grade early renal damage. NGAL levels showed no significant differences. Urinary albumin-creatinine ratio (UACR) did not vary significantly by gender or residency.

Summary: Fluoride is an important trace element in the human body and has beneficial effects when consumed in moderation. However, excessive intake of fluoride can have detrimental effects on various organs and systems. Fluoride pollution has become a significant environmental issue due to the rapid development of agriculture and industry. Despite progress in controlling fluoride pollution, fluorosis still exists. This review summarizes the molecular mechanisms of fluoride toxicity and the pathological changes in organ systems after fluoride poisoning, aiming to provide insights and solutions for the prevention and control of modern fluoride pollution.

Development of a sensitive direct injection LC-MS/MS method for the detection of glyphosate and aminomethylphosphonic acid (AMPA) in hard waters

Summary: Glyphosate, the most widely used herbicide globally, poses challenges for trace analysis in natural waters due to its zwitterionic and polar properties, as well as its ability to complex with cations in the environment. A robust LC-MS/MS method was developed for quantitation of glyphosate and its primary transformation product AMPA in waters of varying hardness, achieving detection limits and consistent accuracy. The method was validated using uncontaminated river water samples from the US and successfully quantified glyphosate and AMPA in surface and drinking water samples from North Carolina and Sri Lanka.

Water sources and kidney function: investigating chronic kidney disease of unknown etiology in a prospective study

Summary: This study reveals that exposure to well water in Wilgamuwa, Sri Lanka is associated with kidney function decline, and identifies a cluster with significantly higher CKDu progression than surrounding areas through geospatial analysis. Analysis of household well samples shows that 68% exceed global water quality standards.

Occupational Paraquat and Glyphosate Exposure May Decline Renal Functions among Rural Farming Communities in Sri Lanka

Summary: The extensive use of herbicides among rural agricultural workers in Sri Lanka, particularly sugarcane farmers, has been postulated to play a role in the development of chronic kidney disease of unknown etiology (CKDu). This study found higher levels of herbicide residues and kidney injury biomarkers among sugarcane farmers in CKDu emerging regions compared to non-endemic areas. These findings suggest a potential link between herbicide exposure and kidney function decline, indicated by markers such as ACR, eGFR, and NGAL.
